    Abstract: In one aspect, the disclosure relates to a method for treating clonal hematopoiesis of indeterminate potential (CHIP) in a subject, the method including at least the step of administering to the subject a therapeutically effective amount of an inhibitor of receptor activator of nuclear factor ?B ligand (RANKL), such as denosumab, a therapeutically effective amount of an anti-inflammatory agent, such as colchicine, or both. In a further aspect, performing the method reduces growth of one or more clones or causes the size of one or more clones to remain constant. In another aspect, each month of performing the method can result in a further reduction of annual clonal growth rate. In a still further aspect, performing the method further treats or prevents a cardiometabolic disease, a cardiovascular disease, or a myeloproliferative disease or disorder. Also disclosed are methods for reducing or maintaining clonal hematopoiesis risk score (CHRS) in a subject.

    Abstract: In one aspect, the disclosure relates to methods for monitoring clonal hematopoiesis of indeterminate potential (CHIP) in a subject, the method including at least performing DNA methylation sequencing on DNA from the subject, performing cell-type deconvolution from the methylation data to estimate a first set of cell-type proportions in the subject, repeating the method to determine a second set of cell-type proportions in the subject, and monitoring a change in variant allele fraction (VAF) using methylation data collected during performance of the method in conjunction with the first set of cell-type proportions and the second set of cell-type proportions. Also disclosed are methods for assessing the performance of a drug for treating a blood cancer.

    Abstract: Compositions and methods are provided for the analysis and treatment of conditions relating to clonal hematopoiesis of indeterminate potential (CHIP). In some embodiments, treatment is provided to reduce the progression of CHIP, particularly to reduce the progression to hematologic malignancy and/or heart disease. In other embodiments, methods are provided for determining clonal expansion, for example as a molecular diagnostic test that enables determination of clonal growth rate from a single sample. The method for determining clonal expansion can be applied to identify factors that influence clonal expansion, including environmental, metabolic, microbiome, and genetic.

    Abstract: Viewing and manipulation systems and methods for medical images shown on a display. The method includes the steps of observing a multiple-person medical environment using a camera having a field of view, and sending field-of-view data of the multiple-person medical environment from the camera to a processor. The processor performs the steps of (i) analyzing the field-of-view data to identify a target practitioner and define a target practitioner-based, non-uniform coordinate frame connected to the target practitioner; (ii) monitoring a time-series of the field-of-view data to identify at least one input communicated by a gesture performed by the target practitioner in the target practitioner-based, non-uniform coordinate frame; and (iii) manipulating a medical image shown by the display in response to identifying the at least one input.
