Abstract: The present invention is directed to synthetic anti-inflammatory peptides and use thereof in the treatment and prevention of inflammatory and fibrotic conditions. Specifically, the invention relates in some embodiments to short isolated peptides having the amino acid sequence Phe-Lys-Glu (FKE), Tyr-Lys-Glu (YKE) or comprising a plurality of these sequences that may be flanked by Ala/Gly (A/G) linkers. The invention further relates in some embodiments to methods for inhibiting scar formation and for treating and alleviating IL-10 dependent conditions.

Abstract: A conditioned cell culture medium of CD11blow human macrophages or a biologically active fraction thereof can be prepared by a method that includes (i) culturing a population of human mononuclear cells of the monocyte/macrophage lineage for 5-7 days, so as to induce differentiation of the mononuclear cells to macrophages; (ii) incubating the macrophages obtained in (i) with apoptotic cells or in the presence of a pro-resolving lipid mediator to reduce the CD11b expression, thus obtaining a culture of CD11blow macrophages; and (iii) collecting the conditioned cell culture medium of CD11blow macrophages. Pharmaceutical compositions containing the CD11blow macrophages conditioned medium or a culture of CD11blow macrophages can be used in the treatment of cancer or fibrosis.

Parent peptides of the sequences Ile-Val-Leu, Glu-Phe-Leu-Asn-Arg-Trp-Ile-Thr (SEQ ID NO:1) and Arg-Met-Leu-Thr (SEQ ID NO:2), were obtained by elastase enzymatic digestion of IL-2, synthesized and modified. The peptides are useful in conditions of acute and chronic inflammation such as in autoimmune diseases.
