    Abstract: Provided are a nanopore preparation and detection method and detection apparatus thereof. The method comprises: forming a nanopore by aggregating a plurality of protein monomers, with a signal detection region of the nanopore being formed in a narrow passage portion of the nanopore; and forming a positive charge cluster in the signal detection region of the nanopore, wherein the charge interaction between the positive charge cluster and negatively charged single molecule analytes that pass through the nanopore can lengthen the residence time of the single molecule analytes in the nanopore. Accordingly, the effective detection of analytes at a single-molecular level is realized, such that single molecule analytes that cannot generate an effective detection signal due to an interaction time with a. nanopore being too short can be effectively detected, the prevalence of single molecule analytes can be significantly improved, and different analytes and detection requirements are accommodated.

    Abstract: Provided is a method for delaminating a composite by immersing the composite into a delamination solution; wherein the composite comprises a metal substrate and a coating applied on one side or both sides of the metal substrate, wherein the coating comprises a polymeric binder; and wherein the polymeric binder comprises an aqueous copolymer. The use of delamination solution comprising a strong base allows for complete delamination of the composite in a highly efficient and extremely fast manner. Furthermore, the delamination method disclosed herein circumvents complex separation processes, contamination and corrosion of the metal substrate and enables an excellent materials recovery. An application of the method for delaminating an electrode for a battery is disclosed herein.

    Abstract: Provided in the present invention is an enzyme reaction solution for constructing a sequencing library and the use thereof. The enzyme reaction solution comprises an enzyme composition and a reaction buffer, wherein the enzyme composition comprises a nucleic acid endonuclease, a DNA polymerase, and a polynucleotide kinase; and the reaction buffer comprises a metal salt, a substrate, and a buffer medium aqueous solution. The present application aims to optimize the formulation of an enzyme reaction solution. The cleavage, terminal repair and addition of A to the terminal of a nucleic acid sample is achieved by a one-step reaction. In a suitable buffer system, the enzyme digestion reaction rate and the terminal repair reaction rate reach a balance. In the case where the initial amount of the sample is 100 pg to 1 µg and the processing time is the same, a sequencing library with a consistent length distribution is obtained.

    Abstract: The present invention provides novel scaffolded HIV-1 vaccine immunogens. Some of the scaffolded immunogens contain a soluble gp140 trimer linked to the N-terminus of the nanoparticle subunit and a T-helper epitope that is fused via a short peptide spacer to the C-terminus of the nanoparticle subunit. Some other immunogens of the invention contain a soluble gp140 trimer protein that is linked to a stable nanoparticle via a short peptide spacer that is a T-helper epitope. Some of the scaffolded immunogens contain a gp140 trimer immunogen presented on a nanoparticle platform formed with I3-01 protein, E2p, or variants of protein 1VLW. Also provided in the invention are nucleic acids that encode the various vaccine immunogens described herein, and expression vectors and host cells harboring the nucleic acids. The invention further provides methods of using the scaffolded HIV-1 vaccine immunogens for preventing or treating HIV infections.
