Publication number: 20210395796Abstract: The present invention relates to a method for amplifying at least one target RNA in a fixed and, optionally, expanded biological sample. In an embodiment of the invention, the method comprises incubating the fixed biological sample with a pair of polynucleotides complementary to non-overlapping and proximal sequences of a target RNA, wherein the polynucleotide pair hybridizes to the target RNA; ligating the polynucleotide pair using a ligase; and amplifying the ligation product. The invention further provides methods for detecting and optionally quantifying and/or sequencing the amplification product. As the method comprises hybridizing polynucleotide pairs to a target RNA in a fixed biological sample, the target RNA can be hybridized in situ.Type: ApplicationFiled: April 30, 2021Publication date: December 23, 2021Inventors: Fei Chen, Asmamaw Wassie, Shahar Alon, Adam Henry Marblestone, Anubhav Sinha, Andrew C. Payne, Edward Stuart Boyden

Patent number: 11180804Abstract: The present invention provides methods for analyzing polynucleotides such as genomic DNA. In some embodiments, the disclosure provides a method for preparing and amplifying a genomic DNA library in situ in a fixed biological sample. The method comprises treating a fixed biological sample with an insertional enzyme complex to produce tagged fragments of genomic DNA. The method further comprises circularizing the tagged fragments of genomic DNA. The method further comprises amplifying the tagged fragments of genomic DNA.Type: GrantFiled: July 24, 2018Date of Patent: November 23, 2021Assignees: Massachusetts Institute of Technology, President and Fellows of Harvard CollegeInventors: Fei Chen, Andrew C. Patent number: 10263942Abstract: Systems and methods for personalizing dialogs are disclosed. A dialog system can provide a user interface to allow a first user to specify a second user to participate in an automated dialog, a specific response option, and a maximum time period for responding. The dialog system can send a message to the second user with the specific response option. The dialog system can branch the dialog based on the occurrence of an event or combination of events occurring in conjunction the second user. In a first branch, the dialog system is configured to send a second communication to the second user using a second communications channel, the second communication containing the specific response option. In a second branch, the dialog system is further configured to execute a third instruction associated with the dialog.Type: GrantFiled: October 19, 2016Date of Patent: April 16, 2019Assignee: Open Text SA ULCInventors: Brian Reistad, William D. Snapper, Andrew C. Payne, James Campbell

Patent number: 9917827Abstract: This invention relates to methods for controlling and monitoring access to network servers. In particular, the process described in the invention includes client-server sessions over the Internet. In this environment, when the user attempts to access an access-controlled file, the server subjects the request to a secondary server which determines whether the client has an authorization or valid account. Upon such verification, the user is provided with a session identification which allows the user to access to the requested file as well as any other files within the present protection domain.Type: GrantFiled: January 9, 2008Date of Patent: March 13, 2018Assignee: Soverain IP, LLCInventors: Thomas Mark Levergood, Lawrence C. Stewart, Stephen Jeffrey Morris, Andrew C. Payne, George Winfield Treese

Patent number: 8635327Abstract: Link traversals leading from an advertisement on the Web to a product page are determined. The determined link traversals are used as a basis for charging for advertising, for example, by counting the number of link traversals from the advertising page to a product page, or by counting the number of sales resulting from such traversals. The effectiveness of advertising on the Web is evaluated by measuring the number of sales or transactions resulting from link traversals from the advertisement to the page.Type: GrantFiled: April 12, 2000Date of Patent: January 21, 2014Assignee: Soverain Software LLCInventors: Thomas Mark Levergood, Lawrence C. Patent number: 8494884Abstract: A system is provided that enables customers to search for event tickets for purchase. A computer interacts with the user to allow the user to specify criteria for event tickets the user wishes to purchase. The system presents relevant event ticket results, and provides filters for the user to further refine those search results. The user selects an event, and then selects tickets for that event. Ticket availability is shown, with seat locations indicated directly on an interactive venue map. For purchase, the user is directed to the selling ticket provider.Type: GrantFiled: March 24, 2011Date of Patent: July 23, 2013Assignee: Nextag, Inc.Inventors: Andrew C.
