    Abstract: A pneumatically controlled aquaculture apparatus is provided. The apparatus has a lift vessel that lifts containers holding shellfish out of the water when the lift vessel is floating in order to allow air desiccation to prevent bio-fouling of the equipment and shellfish. The apparatus has a floatation control tank positioned above the containers holding shellfish in a centered position of the apparatus. The apparatus also has a suspension buoy for suspending the apparatus in the water column. The apparatus may be deployed in two different suspended positions with either the floatation control tank or the suspension buoy floating on the water surface and the containers holding shellfish suspended below the surface. The apparatus may also be deployed in a submerged position in which the apparatus is resting on the water bottom.

The apparatus also has a buoy bladder positioned above the containers holding shellfish. The buoy bladder is retained in a centered position of the apparatus. The apparatus may be deployed in a suspended position with the buoy bladder floating on the water surface and the containers holding shellfish suspended below the buoy bladder. In addition, the apparatus may be deployed in a submerged position in which the buoy bladder is deflated and the apparatus is either resting on the water bottom or retained in a desired position in the water column.

Each tank has an air supply line connected to the tank and an opening on the bottom side of the tank. Each air supply line is connected to a manifold for controlling airflow to the tanks. Air is used to displace water in the tanks by pushing the water out of the openings in the bottom of the tanks in order to float the frame. To submerge the frame, the tanks are depressurized to allow water to displace the air in the tanks. When floating, the tanks lift the frame and the containers out of the water to allow air desiccation in order to prevent bio-fouling of the equipment and shellfish.

    Abstract: Methods and kits for detection of bacteria, especially Vibrio parahaemolyticus and Vibrio vulnificus, are provided using a unique combination of selective ingredients and two-phase culture (solid-phase culture gel and liquid-phase culture/enrichment broth) allows for high sensitivity and specificity of the kits for growth of Vibrio parahaemolyticus and Vibrio vulnificus in the detection methods and kits. The invention, through the detection mechanism accomplished by the novel formulation of selective ingredients and the two-phase culture, allows for real-time detection of a single cell of Vibrio parahaemolyticus and Vibrio vulnificus within 24±2 hours of introducing a target sample to the Vibrio parahaemolyticus and Vibrio vulnificus detection kits.
