  • Publication number: 20040191219
Abstract: Stabilized pharmaceutical compositions comprising substantially monomeric interferon-beta (IFN-&bgr;) and methods useful in their preparation are provided. The compositions comprise the IFN-&bgr; solubilized in a low-ionic-strength formulation that maintains the composition at a pH of about 3.0 to about 5.0. Methods for preparing these compositions, and for increasing solubility of IFN-&bgr; in pharmaceutical compositions, are provided.
    Type: Application
    Filed: April 9, 2004
    Publication date: September 30, 2004
    Applicant: Chiron Corporation
Inventors: Bret A. Shirley, Susan Babuka, Bao-Lu Chen, Maninder Hora, Minna Choe, Melanie Tellers
  • Publication number: 20040180827
Abstract: Lyophilized compositions of TFPI or a TFPI variant suitable for long-term storage can be formed by lyophilizing an aqueous formulation of the TFPI or TFPI variant having a pH from about 4 to about 8 with a carbohydrate or amino acid glass forming agent. These lyophilized compositions are stable for greater than three months when stored at 40° C.

  • Publication number: 20030198602
Abstract: Methods for administering interleukin-2 (IL-2) or variants thereof via pulmonary inhalation are provided. The methods comprise preparing a pharmaceutical composition comprising IL-2 or variants thereof for subsequent delivery as an aqueous or nonaqueous solution or suspension or a dry powder form. These compositions may further comprise a surfactant in an amount sufficient to enhance absorption of the composition following pulmonary inhalation of the composition. When used in the delivery methods of the invention, these highly absorbable compositions result in enhanced bioavailability of IL-2.

  • Publication number: 20030180253
Abstract: Stabilized liquid polypeptide-containing pharmaceutical compositions are provided. The compositions comprise an amino acid base, which serves as the primary stabilizing agent of the polypeptide, and an acid and/or its salt form to buffer the solution within an acceptable pH range for stability of the polypeptide. The compositions are near isotonic. Methods for increasing stability of a polypeptide in a liquid pharmaceutical composition and for increasing storage stability of such a pharmaceutical composition are also provided.
    Type: Application
    Filed: November 18, 2002
    Publication date: September 25, 2003
    Applicant: Chiron Corporation

  • Publication number: 20020137884
Abstract: A method of modifying protein solubility employs polyionic polymers. These facilitate the solubilization, formulation, purification and refolding of proteins especially incorrectly folded proteins and aggregated proteins. Compositions are described that are suitable for formulating TFPI. The compositions allow preparation of pharmaceutically acceptable compositions of TFPI at concentrations above 0.2 mg/mL and above 10 mg/mL.
