    Abstract: Provided herein are systems and methods for the enrichment and detection of nucleic acid molecules. In particular, provided herein are systems and methods for the selective enrichment of a desired nucleic acid molecule in a sample containing non-desired nucleic acid molecules in which a probe with different complementarity to the the desired compared to the undesired nucleic acid, e.g. wild-type and variant. The probe may be bound to the solid phase, e.g. via biotin, the probe binding to the undesired may be cleaved so that the undesired is released and the desired nucleic acid is enriched by binding to the solid phase. The method may also employ differential release from the probe.
