    Abstract: The invention relates to a process for the manufacture of very high-purity antihemophilic factor (FVIIIc) and von Willebrand factor. This process enables the manufacture of very high-purity antihemophilic factor (FIIIc) devoid of the bulk of the Willebrand factor comprises a step for purification by ion exchange chromatography with the aid of a chromatography column containing a gel, the purification step comprising a step for adsorption of the antihemophilic factor essentially devoid of the Willebrand factor on the gel in the column and a step for desorption of the purified antihemophilic factor, which is collected, thereby obtaining an antihemophilic factor devoid of the bulk of the Willebrand factor and having an activity as high as 250 IU/mg of proteins. This process also permits to recover von Willebrand factor in very high purity.

    Abstract: The invention relates to a process for the manufacture of a high-purity activated factor VII concentrate.This process comprises the use of a plasma free of cryoprecipitate, preferably of human origin, as well as at least one purification step involving chromatography at least once on an ion exchange resin, and a factor VII activation step, wherein the first stage is direct activation of the factor VII in the crude supernatant of plasma free of cryoprecipitate, without the addition of exogenous proteins.By virtue of the invention, the high-purity activated factor VII is essentially free of vitamin K-dependent factors and factors VIIIC and VIIICAg and has a factor VIIa/factor VII ratio greater than 5 with a specific activity of the activated factor VII greater than 200 IU/mg of proteins.

This process comprises the selective adsorption of factor VIIIc on the gel of a first ion exchange chromatography column, the von Willebrand factor being contained in the non-retained fraction, and is characterized in that the non-retained fraction is treated so as to adsorb the von Willebrand factor selectively on the gel of a second chromatography column, the factor then being desorbed and being obtained with a very high purity.
