    Abstract: A specific blocking reagent, method of manufacture thereof and methods of use are provided. The blocking reagent uses the specific antibody structure recognized by the target, e.g., analyte, and any interfering substances, e.g., heterophile molecules. The blocking reagent is, however, an inactivated form of the target-specific antibody. Thus, the blocking reagent cannot bind the target because the target-binding regions have been inactivated. The blocking reagent can, however, block interfering substances such as heterophile molecules as these binding regions remain active. In this manner, the blocking reagent provides highly specific removal of interferents. The blocking reagent thus has broad applicability in immunoassays as well as enhanced delivery of therapeutic and/or diagnostic agents.
