    Abstract: The present invention include methods and compositions for treating bone-resorbing diseases or bone resorption related to a pathologic condition generally, including, but not limited to osteoporosis, arthritis, rheumatoid arthritis, cancer metastases to the bone, bone cancer, hypercalcemia, osteolytic lesions with orthopedic implants, Paget's disease, and bone loss associated with hyperparathyroidism.     Abstract: The present invention concerns the use of methods to treat bone loss by inhibiting osteoclast precursor cell fusion. Describe herein is the molecular cloning of a polypeptide encoding a protein termed RAIN. The functions of RAIN include regulating osteoclast differentiation and fusion events. In certain embodiments, compositions comprising polynucleotide and/or polypeptides of RAIN may be used in therapeutic compositions and methods for treating bone loss. In various embodiments, bone loss may be treated by inhibiting the differentiation of osteoclast. Inhibition of osteoclast differentiation may be accomplished by inhibiting osteoclast precursor cell fusion, thus inhibiting the formation of a multi-nucleate resorptive osteoclast.

    Abstract: The present invention provides an isolated and purified protein that associates with the cytoplasmic domain of the p60 form of the tumor necrosis factor receptor, having a molecular weight of about 52-55 kDa on SDS-PAGE, is a phosphoprotein, and does not bind to the p80 form of the tumor necrosis factor receptor. Also provided is an isolated and purified protein kinase that binds to the cytoplasmic domain of the p60 form of the tumor necrosis factor receptor, said kinase phosphorylates the p60 form of the tumor necrosis factor receptor. Also provided are various methods of manipulating this tumor necrosis factor receptor-associated protein and kinase in order to reduce various biological effects of tumor necrosis factor.
