    Abstract: Methods and compositions regarding autocatalytic DNA strand exchange with a pre-defined target DNA are described herein. Specifically, single stranded DNA or double stranded DNA construct comprising preferably four definable sequences facilitate phospho-diesterase hydrolysis reactions and subsequent phosphodiesterification reactions or a transesterification reaction between the construct and a target DNA. The reactions require no mediation by protein catalytic mechanisms and are useful for therapeutic applications regarding genetically related diseases.

    Abstract: An expression vector for altering expression of a target nucleic acid sequence in a host cell by production of single-stranded cDNA (ssDNA) in the host cell in vivo. The expression vector is comprised of a cassette comprising a sequence of interest, an inverted tandem repeat, and a primer binding site 3′ to the inverted tandem repeat, and a reverse transcriptase/RNAse H coding gene, and may be transfected into the host cell. Transcription of the cassette by the host cell produces an RNA template which is reverse transcribed with the product of the RT coding gene to produce ssDNA of a specified sequence. The ssDNA is modified to remove flanking vector sequences by taking advantage of the “stem-loop” structure of the ssDNA, which forms as a result of the inverted tandem repeat that allows the ssDNA to fold back on itself, forming a double stranded DNA stem.

    Abstract: Methods and compositions for the isolation and specific targeting of any single stranded DNA sequence to be acted upon by any desired double stranded DNA genetic element or recognition sequence in a cis-oriented fashion. The invention involves construction of a vector comprised of single stranded sequences that form "stem-loop" structures wherein the "loop" comprises the single stranded target sequence and the "stem" comprises the double stranded "functional" cis-acting genetic elements or recognition sequences. The in vivo formation of this single stranded intermediate (prior to stem-loop folding) is performed by genetic elements which direct the normal DNA replication of any one of a number of prokaryotic and eukaryotic viruses during their life cycles. Constructs containing these replicative functions are used to produce the desired single stranded intermediates. Also included in the stem-loop structure is an appropriate inverted tandem repeat which, forms the "stem" of the stem-loop structure.
