    Abstract: An operation method of a multiplex slide plate device is provided. First, the multiplex slide plate device is assembled, including a slide plate, a sacrificial layer and a housing. The slide plate has reaction vessels, and the sacrificial layer has a microfluidic channel composed of an injection channel, a main channel and a distal channel. A sample solution is injected to the injection channel, such that the sample solution flows from the injection channel through the main channel to the distal channel, wherein the sample solution loads into the reaction vessels. Afterwards, an oil is injected to the injection channel, such that the oil flows from the injection channel through the main channel to the distal channel, wherein the oil removes the sample solution not loaded into the reaction vessels. Next, the sacrificial layer is heated to melt, and the melted sacrificial layer is mixed with the oil.

Excess waste liquid flows from the exhaust hole into the storage tank, and is covered by the oil and cannot reflow.

    Abstract: A measuring method for low concentration nucleic acid sample is provided. The measuring method is a nucleic acid measuring method for samples of low nucleic acid concentration range in a qPCR experiment, so as to extend the linear dynamic range of experimental detection and increase detecting sensitivity by correcting the original Cq value. The method includes providing a test plate with a plurality of reaction wells or holes, so as to perform qPCR reaction on the nucleic acid samples. Next, performing the adjustment step based on the positive well measurement value derived from the number of positive wells, so as to correct the Cq value of qPCR reaction to the expected value of linear range.

    Abstract: The microarray biochemical reaction device of this invention integrates microfluidic trenches with a microarray to form a serpentine microchannel passing through all DNA probes provided in the microarray. A sample solution is introduced into the microchannel and scrambled into discrete plugs to induce droplet mixing. The discrete plugs are then shuttled through the entire microchannel (shuttle hybridization), sweeping over DNA probes to perform hybridization. Using chaotic mixing of droplets, the hybridization efficiency is enhanced and reaction time for hybridization is shortened. During shuttling, the plugs are thoroughly mixed by the natural re-circulating flows. Method for the preparation of the microarray biochemical reaction device is also disclosed.

    Abstract: The present invention discloses a method for preparation and surface modification of plastic microfluidic chip is disclosed. A microfluidic chip is first prepared by micromachining using a laser scriber on a plastic substrate, such as a PMMA substrate, to have a desired trench width or trench widths and desired aspect ratio. Surface modification is obtained after a thermal annealing treatment, followed by a chemical modification process. During the chemical modification diverse functional groups such as perfluoroalkyl (—CnF2n+2), amino (—NH2) or sulfhydryl (—SH) are introduced to perform surface passivation or further biomolecules immobilization. A plastic microfluidic chip with modified surface characters is thus obtained. The present invention also discloses plastic microfluidic chips so prepared.
