    Abstract: This invention provides populations human cells of the cardiomyocyte lineage. The cells are obtained by causing cultures of pluripotent stem cells to differentiate in vitro, and then harvesting cells with certain phenotypic features. Differentiated cells bear cell surface and morphologic markers characteristic of cardiomyocytes, and a proportion of them undergo spontaneous periodic contraction. Highly enriched populations of cardiomyocytes and their replicating precursors can be obtained, suitable for use in a variety of applications, such as drug screening and therapy for cardiac disease.

Culturing human embryonic stem cells in fresh medium according to this invention causes the cells to expand surprisingly rapidly, while retaining the ability to differentiate into cells representing all three embryonic germ layers. This new culture system allows for bulk proliferation of pPS cells for commercial production of important products for use in drug screening and human therapy.

    Abstract: Disclosed is a technique for compensating for optical passband shift of an optical component (for example an optical demultiplexer). A broadband source coupled to a dispersive element generates a chirped pulsed optical signal. Data is modulated onto particular wavelengths of the chirped pulsed optical signal by appropriately synchronizing a data modulator. The modulated signal is transmitted to the downstream optical demultiplexer, which may be subject to passband shift due to, for example, changes in environmental conditions. A feedback signal from an output port of the demultiplexer is provided to the transmitter and is used to phase shift the modulator. The phase shift results in effectively adjusting the wavelength onto which the data is modulated to substantially correspond to the passband centers of the demultiplexer.

    Abstract: An optical device including a microstructured fiber pumped by an external pulsed-light source. In one embodiment, the microstructured fiber includes two waist regions functioning as a tunable attenuator and a wavelength shifter, respectively. Output wavelength of the optical device is selected by attenuating the pump light in the first waist region and then passing the light through the second waist region to shift the pump energy to a new spectral band. An optical device of the invention configured with two or more microstructured fibers generates two or more synchronized pulsed beams, each at a different characteristic wavelength. Certain embodiments of the invention provide an inexpensive, compact, energy-efficient multi-wavelength synchronized pulsed-light source.

    Abstract: Apparatus and method is described for using a silicon photon-counting avalanche photodiode (APD) to detect at least two-photon absorption (TPA) of an optical signal, the optical signal having a wavelength range extending from 1.2 &mgr;m to an upper wavelength region that increases as the number of photons simultaneously absorbed by the APD increases beyond two. In one embodiment, the TPA count is used by a signal compensation apparatus to reduce dispersion of a received optical pulse communication signal subjected to group velocity dispersion, polarization mode dispersion, or other signal impairment phenomena which effect the TPA count. Another embodiment, the TPA count is used to determine the optical signal-to-noise ratio of a received optical pulse communication signal. Another embodiment uses the TPA count to determine the autocorrelation between a first and second optical pulse signals as a function of the relative delay between the first and second optical pulse signals.

    Abstract: This disclosure provides a system for obtaining expression libraries from primate pluripotent stem (pPS) cells. pPS cells can be maintained in vitro without requiring a layer of feeder cells to inhibit differentiation. The role of the feeder cells is replaced by several other culture conditions provided in a suitable combination. Conditions that promote pPS cell growth without differentiation include supporting the culture on an extracellular matrix, and culturing the cells in a medium conditioned by another cell type. The cDNA libraries from such cultures are devoid of transcripts of feeder cell origin, relatively uncontaminated by transcripts from differentiated cells, and can have a high proportion of full-length transcripts. Subtraction libraries can also be produced that are enriched for transcripts modulated during differentiation.

    Abstract: The invention involves providing a microstructured fiber having a core region, a cladding region, and one or more axially oriented elements (e.g., capillary air holes) in the cladding region. A portion of the microstructured fiber is then treated, e.g., by heating and stretching the fiber, such that at least one feature of the fiber microstructure is modified along the propagation direction, e.g., the outer diameter of the fiber gets smaller, the axially oriented elements get smaller, or the axially oriented elements collapse. The treatment is selected to provide a resultant fiber length that exhibits particular properties, e.g., mode contraction leading to soliton generation, or mode expansion. Advantageously, the overall fiber length is designed to readily couple to a standard transmission fiber, i.e., the core sizes at the ends of the length are similar to a standard fiber, which allows efficient coupling of light into the microstructured fiber length.

    Abstract: This invention provides media that support the growth of primate pluripotent stem cells in feeder-free culture, and cell lines useful for producing such media and other purposes. Conventionally, it has been necessary to grow pluripotent embryonic cells on feeder layers of primary embryonic fibroblasts, in order to prevent them from differentiating. It has now been discovered that standard culture media conditioned by special cell lines can be used to support proliferation of pluripotent stem cells while inhibiting differentiation in an environment free of feeder cells. This invention includes mesenchymal and fibroblast-like cell lines obtained from embryonic tissue or differentiated from embryonic stem cells. Methods for deriving such cell lines, processing media, and growing stem cells using the conditioned media are described and illustrated in this disclosure.

The N-photon absorption rate can also be used to determine the optical signal-to-noise ratio of a received optical pulse communication signal, and/or to synchronize a second optical pulse signal with the first optical signal.

    Abstract: A high bit rate, long haul optical communication system encodes a polarization interleaved stream of RZ optical pulses using phase shift keying (PSK) or differential phase shift keying (DPSK), in contrast to conventional on-off keying (OOK). The polarization interleaved stream of RZ optical pulses can be used for PSK or DPSK encoding of either one data stream having a bit rate that is the same as the optical stream pulse rate, or two (or more) independent data streams which individually each have lower bit rates, but which, when combined, have the same rate as the optical stream pulse rate. The latter arrangement essentially accomplishes polarization multiplexing (P-MUX). Individual wavelengths can be combined in a WDM or DWDM system, wherein, at the transmitter, multiple streams of polarization interleaved pulses, each stream having a different wavelength, are combined.
