    Abstract: A method for determining a total number of fibers in a muscle tissue based on panoramic scanning is described. A muscle tissue slide is panoramically scanned to obtain a panoramic digital slide using a panoramic scanner, and the panoramic digital slide obtained is segmented, counted and summed to realize the statistical effect on the total number of fibers in the muscle tissue; compared with dependence on an optical microscope, 5-10 fields of view of an image are represented as histological characteristics of the muscle fiber of an biological individual, and further compared with a method using a product of average muscle fiber density and muscle slide area as an estimated value of the total number of fibers of the slide, the present disclosure can realize full-field statistics of the muscle tissue, with a smaller error and a more accurate result.

    Abstract: A method for generating a genome-edited chicken is provided, relating to the technical field of genetic engineering. Ovarian injection in situ is conducted on a hen, where a gene editing reagent is injected into ovarian medulla of the hen that is close to laying eggs, such that the exogenously-injected gene editing reagent can enter developing ovarian follicles through blood circulation. In resulting Go individuals, a chimera chicken with both somatic cells and germ cells edited is successfully and efficiently obtained, with an editing efficiency of the Go individuals reaching up to 36.36%. Compared with a traditional primordial germ cell (PGC)-mediated method, the ovarian injection in situ is time-saving and labor-saving, convenient and rapid, low-cost, and highly safe.

The SNP loci of the chip are respectively derived from 14,624 SNP loci shared by each line of the major egg-type chickens in China; 3,677 SNP loci associated with disease-resistant traits in egg-type chickens; 16,000 SNP loci associated with economic traits in egg-type chickens; and 9,358 SNP loci making up genomic regions not covered by the foregoing probes. The 43,681 SNPs on the egg-type chicken whole-genome SNP chip have DNA sequences as represented by SEQ ID NO. 1-43,681. The chip can specifically identify the genetic relationship between commercial egg-type chickens and egg-type chickens of local breeds, and can also perform applications such as whole-genome association analysis, genome selective breeding, and QTL location analysis of target traits and population genetic analysis.
