  • Patent number: 12359248
Abstract: Described herein are synthetic methods for producing sequence-specific RNA oligonucleotides that eliminate impurities produced in prior art methods. In one aspect, a first amplification primer includes one or more deoxyuridine residues, wherein at least one of the one or more deoxyuridine residues is at position ?1, ?2, ?3, ?4 or ?5 of the promoter region for the single-subunit, DNA-dependent RNA polymerase. The deoxyuridines are excised to provide an amplified functional template DNA which is then used to synthesize RNA which has reduced immunogenic double stranded RNA compared to controls.

  • Patent number: 12330256
Abstract: A multi-tool positioning and manufacturing system moves a workpiece among many tools, such as grinding wheels, each of which performs a manufacturing operation on the workpiece. The system moves the workpiece automatically, quickly, repeatably, and precisely among the tools, without any manual intervention. It can perform the same manufacturing operations as many separate tools, such as grinding the inner diameter, outer diameter, and rib of a tapered roller bearing cone. Because a machinist does not have to move the workpiece between machines for different operations, the total manufacturing process can be faster and higher yield than with separate tools. The system can also move a single dresser among the tools for dressing and truing, further increasing manufacturing efficiency by eliminating the need for separate dressers for separate tools.

  • Patent number: 12325873
Abstract: Described herein are synthetic methods for producing sequence-specific RNA oligonucleotides that eliminate impurities produced in prior art methods. In one aspect, an end-protected capture DNA complementary to a portion of the product RNA is employed. In another aspect, the template DNA is covalently or noncovalently linked to the RNA polymerase, either directly or through the use of a nontemplate DNA. In a third aspect, a flow chamber is employed. All of the methods can be used in combination.

  • Patent number: 12313052
Abstract: A progressive sealing system for a reciprocating machine includes a pressure breaker having a first seal, and one or more additional seals between the pressure breaker and an end plate. One or more seal housings hold the seals between the first seal and the end plate. The end plate couples the progressive sealing system to a housing of a cylinder of the reciprocating machine such that a surface of the first seal faces a chamber of the cylinder. A housing for the second one of the seals includes a bore having an inner surface. The inner surface of the bore of the second seal housing and an outer surface of a rod of the reciprocating machine define an annular space when the rod is received in the bore. The packing case includes an expansion chamber adjoining the annular space.
