    Abstract: The present invention relates to methods for identifying interacting regions of transcription factors, and methods for identifying agents which modulate the interactions, useful for affecting gene regulation, for example, cellular transformation. A site within residues 130–154 and within residues 343–358 in Stat3 were found to interact with the transcription factor c-Jun. On c-Jun, a site within residues 105 and 334, and more particularly, between 105 and 263, interact with Stat3. These sites of interactions permit methods for identifying agents which modulate the interaction between these transcription factors to modulate gene transcription.

    Abstract: The invention relates to compositions and methods for modulating cell signaling mediated by signal transducers and activators of transcription (STAT). The compositions target cellular STAT3 and STAT1 protein, particularly STAT3, for degradation via the ubiquitination pathway. Thus, the STAT inhibiting agents are useful for inhibiting STAT mediated signal transduction events, such as responses to IL6 and v-Src, any may be applied to treating diseases associated with activated STAT proteins, particularly STAT3 activity, such as cell proliferative disorders, inflammatory reactions, and autoimmune conditions.

    Abstract: New hybrid or chimeric fusion polypeptide transcription regulators which induce interferon target gene expression are provided for use in anti-viral and anti-proliferative applications. The hybrid fusion proteins comprise the p48 interferon regulatory factor protein, or a functional portion thereof, fused to a transcriptional activation domain (TAD), or a functional portion thereof. More specifically, the p48 interferon regulatory factor protein is fused to a STAT transcriptional activation do main (TAD) (p48-STAT TAD). A particular p48-STAT TAD is p48-S2C, in which S2C is the TAD of STAT2. Another p48-TAD is p48-VP16 TAD, in which p48 is fused to the transcriptional activation domain of the VP16 protein of Herpes Simplex Virus (HSV). The p48-TAD nucleic acid sequences and encoded polypeptides can be employed for anti-viral treatment and for the treatment of cancers, tumors and neoplastic diseases.
