The invention uses CRISPR/Cas9 technology to realize site-directed editing rice RAY1 gene, through knocking out rice RAY1 gene by frameshift mutation, the protein RAY1 is inactivated, and a new generation of rice germplasm with significantly improved yield is obtained.

    Abstract: A high-throughput and rapid nucleic acids detection method based on capillary microarrays comprises the steps that firstly, microarray containing a number of hydrophilic and vertical micro-channels is fabricated by capillary assembling, casting and machining, and the outer surface of the capillary array is coated with super-hydrophobic Ultra-Ever Dry paint; secondly, different primer sets are individually loaded into the micro-channels and air-dried to adhere them on the inner surface, and then the microarray is anchored into a reaction tube; thirdly, the reaction mixture is introduced into every microchannel at once through capillary force by a special designed sample-loading adaptor, and then the amplification reaction is performed in the temperature control device; and finally, the fluorescence can either be measured continually during the reaction for real-time detection or be recorded once in the end for endpoint detection. Moreover, the products can also be recovered for other use later.

    Abstract: Provided is a breeding method for two lines hybrid rice based on the rice mutant osms4 (Oryza sativa male sterile 4), which was originally selected from the mutagenizing seeds of japonica cultivar 9522 with 60Co gamma ray, and the stable inheritance osms4 mutant was obtained after three generations backcrossing, which is regulated by a recessive single gene OsMS4. The invention is related to the usage of OsMS4 gene (also known as Carbon Starved Anther, the CSA gene), osms4 mutant can be used as male sterile line under short day condition in both japonica and indica background, which grows abnormal anther with defection during second mitosis stage and produces completely sterile pollen. While under the long day condition, the osms4 mutant can be used as maintainer liner and produces normal pollen.
