    Abstract: Provided are functional segregated telodendrimers having, for example, two or three functional segments. The telodendrimers can aggregate to form nanocarriers. The telodendrimers can have one or more tetracycline drugs (such as CMT-3) physically bound thereto that release under physiological conditions. Such nanocarriers with loaded CMT-3 disperse stably in aqueous solution and may be used in treating inflammatory disease through local or systemic administration. Also provided are methods of treating acute respiratory distress syndrome (ARDS) and/or CARDS in a subject in need thereof, including: administering a therapeutically effective amount of CMT-3 and one or more nonantimicrobial host-modulators directly to the lungs of the patient. In embodiments, CMT-3 is administered to the lungs in an aerosolized formulation. In embodiments, subsequent to CMT-3 administration, one or more non-antimicrobial host-modulators directly to the lungs of the patient.
