    Abstract: The invention provides methods and reagents for identifying an agent, such as by screening a library of agents, that modulates the interaction of two or more polypeptides, the method comprising: introducing into a cell at least a first polypeptide, each comprising a binding domain, wherein the first polypeptide comprises a localization domain of the second polypeptide; and detecting the cellular location of the first polypeptide, the second polypeptide or a combination thereof, wherein a change in the cellular location of the first polypeptide, the second polypeptide or a combination thereof indicates that the agent modulates the interaction of the two or more polypeptides. The invention also provides methods and reagents for identifying the binding domains of one or more polypeptides.
