    Abstract: A system and method for maintaining the vitality of an organ through negative pressure ventilation and perfusion. The system includes fluidically coupled components: an organ enclosure, a diaphragm enclosure, an actuator/pump, a perfusion system, and a reservoir. The actuator can displace a precise amount of a working fluid that displaces that precise amount of a sterile support fluid. The sterile fluid travels between the diaphragm enclosure and the organ enclosure, thereby ventilating the organ within the organ chamber. The perfusion system circulates a perfusate through the organ.

    Abstract: This disclosure relates to a gravity-driven infusion system that uses image-based monitoring to regulate flow. A contrasting, infrared-backlit wall and camera capture pendant drops and meniscus levels within a transparent drip chamber. The controller defines a baseline referenced to the spout or meniscus and fits sparse spline points on the drop perimeter to derive geometric functionals, such as neck width and centroid height. Temporal changes of these functionals map directly to instantaneous flow without explicit volume integration. An optional Young-Laplace model provides a physics-based boundary and confidence metric. A meniscus trend yields a low-frequency flow estimate. Confidence-weighted fusion controls a flow-control valve that compresses a multi-lumen insert. An independent safety occluder and watchdog ensure fail-safe shutdown. A medication library stores fluid-aware calibration. Multi-source embodiments orchestrate multiple controllers with virtual head-height equalization and verified handoffs.

    Abstract: Improvements in fluid volume measurement systems are disclosed for a pneumatically actuated diaphragm pump in general, and a peritoneal dialysis cycler using a pump cassette in particular. Pump fluid volume measurements are based on pressure measurements in a pump control chamber and a reference chamber in a two-chamber model, with different sections of an apparatus being modeled using a combination of adiabatic, isothermal and polytropic processes. Real time or instantaneous fluid flow measurements in a pump chamber of the diaphragm pump are also disclosed, in this case using a one-chamber ideal gas model and using a high speed processor to obtain and process pump control chamber pressures during fluid flow into or out of the pump chamber.

    Abstract: A pump cassette comprising an outer flexible membrane covering flowpaths, valve chambers and pump chambers of the cassette is designed to be actuated by a control gasket on a base unit arranged to move designated valve and pump portions of the cassette membrane. The gasket valve control or actuation region is at least partially bounded by a vacuum channel facing the outside of the gasket so that a constant vacuum can be applied between the gasket valve control or actuation region and the adjacent portion of the cassette membrane. An improved version of the vacuum channel comprises a flexible inner wall of the vacuum channel flexes or partially collapses away from the cassette valve seat, while still maintaining patency of the vacuum channel during the application of negative pressure on the gasket valve actuation region to open the cassette valve.

The performance of a cassette valve may improved by optimizing the configuration of the valve control region of the control gasket overlying the cassette valve. This may improve both fluid flow through the valve and reduce the amount of vibratory noise associated with opening the valve. The gasket valve control or actuation region is at least partially bounded by a vacuum channel facing the outside of the gasket so that a constant vacuum can be applied between the gasket valve control or actuation region and the adjacent portion of the cassette membrane.
