    Abstract: The present invention relates to a method for the reduction of repetitive sequences (or the improvement in low-copy sequences) in DNA samples by a combination of restriction endonuclease treatment, followed by adapter ligation, renaturation kinetics-based fractionation, optionally coupled with duplex sequence nucleases and further restriction endonuclease treatment, followed by adapter ligation. The low-copy enriched fractions can be used in further DNA analysis.
