    Abstract: Provided are methods for the diagnosis of allergic disease wherein IgE specific for an allergen of interest is detected in a patient serum sample by using the patient serum sample to sensitize in the presence or absence of an IgE antagonist a mast cell or basophil host genetically engineered to display surface expression of a Fc.epsilon.RI subunit that is capable of mediating the host cells release of a pharmacological mediator upon induction with patient serum and allergen, challenging the sensitized host cells with the allergen of interest, and determining the presence or absence of IgE specific to the allergen of interest in the patient serum sample by comparing the release of the pharmacological mediator produced by host cells sensitized with patient serum in the presence of the IgE antagonist to the release of the pharmacological mediator produced by host cells sensitized with patient serum in the absence of the IgE antagonist.
