Publication number: 20240125776Abstract: Provided herein are encoded microcarriers for analyte detection in multiplex assays. The microcarriers are encoded with an analog code for identification and comprise a capture agent for analyte detection and a substantially transparent magnetic polymer. The analog code is generated by a two-dimensional shape of a substantially non-transparent layer. Also provided are methods of making the encoded microcarriers disclosed herein. Further provided are methods and kits for conducting a multiplex assay using the microcarriers described herein.Type: ApplicationFiled: November 21, 2023Publication date: April 18, 2024Applicant: Plexbio Co., Ltd.Inventors: Dean TSAO, Chin-Shiou HUANG, Cheng-Tse LIN, Chien-Te WU, FengKan LU

Publication number: 20230052147Abstract: Provided herein are methods and kits for detecting the presence of DNA and/or RNA mutations associated with cancer (e.g., lung cancer). The methods and kits employ microcarriers, each with a probe specific for a DNA or RNA mutation and an identifier unique to the probe sequence. Upon isolation and amplification of nucleic acids from a sample, hybridization of amplified DNA with a probe, specific for a DNA or RNA mutation, that is coupled to a microcarrier indicates the presence of the mutation in the sample. Since each microcarrier can be identified through detection of the identifier, multiplex screening assays are provided. Representative genes that can be screened for mutations include, e.g., KRAS, NRAS, PIK3CA, BRAF, EGFR, AKT1, MEK1, and HER2 for DNA mutations and/or ALK, ROS, RET, NTRK1, and cMET for RNA mutations.Type: ApplicationFiled: January 7, 2021Publication date: February 16, 2023Applicant: PLEXBIO CO., LTD.Inventors: Dean TSAO, Chin-Shiou HUANG, Shian Pin HU, Fei-Yu CHANG, Yi Ling HSIEH

Patent number: 10894975Abstract: Provided herein are methods and kits for detecting the presence of DNA mutations in the KRAS, BRAF, CTNNB1, and APC genes. The methods and kits employ microcarriers, each with a probe specific for a DNA mutation in the KRAS, BRAF, CTNNB1, or APC gene and an identifier unique to the probe sequence. Upon isolation and amplification of DNA from a sample, hybridization of amplified DNA with a probe, specific for a DNA mutation, that is coupled to a microcarrier indicates the presence of the DNA mutation in the sample. Since each microcarrier can be identified through detection of the identifier, multiplex screening assays for multiple mutations in each of the KRAS, BRAF, CTNNB1, and APC genes are provided.Type: GrantFiled: December 11, 2017Date of Patent: January 19, 2021Assignee: PLEXBIO CO., LTD.Inventors: Dean Tsao, Chin-Shiou Huang, Shian pin Hu

Patent number: 5869274Abstract: The embodiments of the invention are multiple antibody immunoassay systems, and preferably immunohistochemical staining systems. In these systems the detection antibody system that detects the association of a targeting antibody (that binds to the target moiety to be detected) may also cross-react with endogenous immunoglobulins in the specimen in which the target moiety is to be detected, yielding an unreliable assay with high background levels. This problem has been overcome by a process that includes cross-linking a monovalent antibody to the endogenous antigenic determinants prior to exposure to the targeting antibody and detecting antibody system. The embodiments of the invention also include kits for use in these multiple antibody immunoassay and histochemical staining systems.Type: GrantFiled: July 10, 1997Date of Patent: February 9, 1999Assignee: Zymed Laboratories, Inc.Inventors: Dean Tsao, Zuo-Rong Shi, Peter Luu
