    Abstract: Disclosed are methods of determining methylation status of a target nucleic acid sequence in a sample comprising determining quantitative methylation data at two or more predetermined methylation sites in the target nucleic acid sequence, wherein the target nucleic acid sequence comprises an O6-methylguanine-DNA-methyltransferase (MGMT) promoter. Disclosed are methods of treating a subject having cancer comprising determining, from a sample, quantitative methylation data at two or more predetermined methylation sites in a MGMT promoter, wherein the sample is from the subject having cancer; and treating the subject for cancer when the quantitative methylation data at two or more predetermined methylation sites is at or above a threshold.
