Publication number: 20120177669Abstract: We characterized a total of 175 HLA-DR-associated phosphopeptides using sequential affinity isolation, biochemical enrichment, mass spectrometric sequencing and comparative analysis. Many were derived from source proteins which may have roles in cancer development, growth and metastasis. Most were expressed exclusively by either melanomas or transformed B cells, suggesting the potential to define cell type-specific phosphatome “fingerprints”. We generated HLA-DR?1*0101-restricted CD4+ T cells specific for a phospho-MART-1 peptide identified in two melanoma cell lines. These T cells showed specificity for phosphopeptide-pulsed antigen presenting cells as well as for intact melanoma cells. MHC II-restricted phosphopeptides recognizable by human CD4+ T cells are potential targets for cancer immunotherapy.Type: ApplicationFiled: May 4, 2010Publication date: July 12, 2012Applicants: UNIVERSITY OF VIRGINIA PATENT FOUNDATION, THE JOHNS HOPKINS UNIVERSITYInventors: Suzanne L. Publication number: 20080044915Abstract: The present invention relates to a new method for identifying polypeptides by deducing the amino acid sequence of the carboxy and amino termini by a mass spectrometer analysis. The method comprises the steps of dissociating highly charged peptide precursor ions (e.g., z>4) using electron transfer dissociation inducing anions followed by removal of those reagents and introduction of a second, proton transfer inducing anion type. The second PTR reaction duration is adjusted to convert the ETD products to primarily the +1 charge-state to reduce the highly charged c and z-type fragments, producing an m/z spectrum containing a series of c and z-type fragment ions that are easily interpreted to reveal the sequence of the amino and carboxy terminus, respectively.Type: ApplicationFiled: October 7, 2005Publication date: February 21, 2008Inventors: Donald Hunt, Joshua Coon, John Syka

Publication number: 20060263323Abstract: An alcohol based hand surgical scrub, which includes cationic anti-microbial agent preservatives, cationic polymer film-forming agents and a skin emollient, and provides a long term residual, anti-microbial “invisible glove” on the skin. The composition provides an immediate reduction in bacterial microbes. The polymer system creates an invisible film on the skin following solvent evaporation. This invisible film provides a lasting anti-microbial barrier on the skin which acts to prevent microbial growth.Type: ApplicationFiled: March 24, 2004Publication date: November 23, 2006Applicant: BECTON, DICKINSON AND COMPANYInventors: Minh Hoang, Donald Hunt

Publication number: 20060159690Abstract: The present invention relates to a peptide which is immunologically recognizable as a T cell epitope of the minor Histocompatibility antigen H-Y. The peptide comprises amino acid sequence SPSVDKARAEL (SEQ ID NO: 1) or FIDSYICQV (SEQ ID NO: 2). The peptide is obtainable from the minor Histocompatibility antigen H-Y. Providing a toxic moiety to the peptide eliminates T cells having specific binding affinity for the peptide. The peptide induces tolerance for transplantations when administered to H-Y-negative recipients.Type: ApplicationFiled: December 16, 2005Publication date: July 20, 2006Inventors: Els Goulmy, Donald Hunt, Victor Engelhard

Publication number: 20060073517Abstract: The present invention relates to the generation of antibodies that bind to specific modifications of the amino terminus of histone H3 and the carboxy terminus of the H2A and H2B peptides. More particularly, the present invention is directed to the generation of a set of antibodies that recognize various post-translational modifications of histone proteins that function as switches for regulating transcription and mitosis. Compositions comprising these antibodies are used as diagnostic and screening tools.Type: ApplicationFiled: March 9, 2004Publication date: April 6, 2006Inventors: C Allis, Wolfgang Fishle, Yanming Wang, Donald Hunt

Publication number: 20050277161Abstract: The present invention describes novel tumor-specific phosphorylated peptides, nucleic acids encoding those peptides, and antibodies generated against said peptides. The genes, peptides, and antibodies described herein may be used as diagnostic indicators of the presence of cancer and/or used in therapeutics to treat cancer.Type: ApplicationFiled: June 7, 2005Publication date: December 15, 2005Inventors: Victor Engelhard, Angela Zarling, Donald Hunt, Anne Evans, Jeffrey Shabanowitz, Joy Polefrone, Leann Hopkins

Publication number: 20050199804Abstract: The present invention relates to a new method for fragmenting ions in a mass spectrometer through the use of electron transfer dissociation, and for performing sequence analysis of peptides and proteins by mass spectrometry. In the case of peptides, the invention promotes fragmentation along the peptide backbone and makes it possible to deduce the amino acid sequence of the sample, including modified amino acid residues, through the use of an RF field device.Type: ApplicationFiled: March 14, 2005Publication date: September 15, 2005Inventors: Donald Hunt, Joshua Coon, John Syka, Jarrod Marto
