    Abstract: The disclosure provides methods, systems and related software, for automated or semi-automated sorting and/or isolating cells with visually distinguishable phenotypes. In some embodiments, the methods comprise providing a plurality cells with a photo-activatable detectable marker in their respective nuclei. The plurality of cells are imaged and, based on the image, the status for one or more visually identifiable phenotypes are determined. Cells determined to have the desired phenotype status are specifically exposed to a light wavelength for a time sufficient to uniquely activate the photo-activatable detectable marker in the individual cells with the desired phenotype status. The cells are then sorted on the basis of the activated detectable marker. The disclosure also provides methods for preparation and isolation of nuclei from fixed, adherent cells for analysis.

    Abstract: The disclosure provides compositions and methods for suppressing off-target editing guide RNA-nuclease complexes. The disclosed strategies incorporate use of catalytically inactive truncated guide RNA/nuclease complexes to shield off-target editing. In some embodiments, the disclosure provides a method of inhibiting off-target cleavage of DNA by a first guide RNA-endonuclease complex by contacting the DNA with a second guide RNA-endonuclease complex that comprises a second guide RNA corresponding to the off-target site but with a recognition sequence of 16 or fewer nucleotides. In another aspect, the disclosure provides a method for preventing cleavage of DNA after editing and subsequent homology-directed repair (HDR) by contacting the repaired DNA with a guide RNA-endonuclease complex that comprises a guide RNA with a guide RNA corresponding to the repaired sequence but with a recognition sequence of 16 or fewer nucleotides. Additional methods, compositions, and kits are also provided.

The device essentially comprises two triangular support frames and an attached stabilizer bar, which when assembled yields a pyramidal prismatic structure upon which the user performs a variety of exercises. The configuration of the device changes as the user optionally places an exercise grip bar in various receiving grooves. This device is believed to be useful in the home setting, whereby the individual user may obtain a full work-out without the need for larger home gym systems. A device according to the invention is also believed to be favorable for use in large gym environments, hospital therapy rooms, classes and other fitness-related facilities.
