Abstract: Provided herein are improved methods of producing therapeutic circular DNA vectors, pharmaceutical compositions produced by such methods, and methods of using pharmaceutical compositions. The invention is based, at least in part, on cell-free manufacturing processes involving restriction digest and ligation schemes, such as restriction digest processes involving type IIs restriction enzymes. Methods provided herein are amenable to large scale production of high-purity compositions of therapeutic circular DNA vectors.
