  • Patent number: 5864021
Abstract: Transforming growth factor .beta. (TGF-.beta.) is produced in relatively large quantities and at a relatively high purity by fermentation in a perfusion microcarrier reactor. Conditioned media from the reactor is first treated to provide the active form of TGF-.beta. and subsequently purified by cation exchange chromatography followed by hydrophobic interaction chromatography. Optionally, nucleic acids complexed with the TGF-.beta. may be removed while the protein is bound to the cation exchange resin. Purified TGF-.beta.1 compositions have a specific activity above 10.sup.7 U/ml.

  • Patent number: 5378816
Abstract: Proteins having an epidermal growth factor-like binding region, such as thrombomodulin, may be recovered and purified on a anion exchange resin, such as a polyethyleneimine resin. Conditioned medium containing recombinantly produced protein is applied to the resin under conditions of pH and ionic strength selected to result in specific binding of the protein to the resin. The column is then washed to remove non-bound proteins and bound proteins other than the desired protein to be separated. The desired protein is then eluted from the column and collected in a medium. Further purification, concentration, and other treatment of the protein may be performed by conventional techniques.

  • Patent number: 4303740
Abstract: The invention relates to a new process for the preparation of highly accurate and stable silver-silver salt reference electrode. According to the invention a homogeneous mixture is formed from silver powder and a powdered silver salt, then a paste is prepared by blending the mixture with a liquid binding agent capable of adhering to metals, applying the paste onto the surface of an electrode base, allowing the paste to harden and then subjecting the electrode to a heat treatment between 100.degree. C. and 250.degree. C. In this latter step a porous silver-silver salt layer is formed on the metal base, which renders the electrode highly accurate and stable.Ion selective measuring electrodes are prepared essentially in the same manner with the difference that silver is replaced partly or entirely by graphit.

  • Patent number: 4120657
Abstract: The invention relates to a process of and equipment for a selective, quick and accurate analysis of liquid samples. According to the process of the invention a liquid or a gas sample absorbed by a liquid is allowed to flow at a constant volume rate and overtitrated with an amount of a reagent adequate to the measurement in a definite time. Then the flow of the reagent is decreased to zero in a definite time, meanwhile continuously following, in a known way, the changes occurring in the reaction mixture. The determination of the concentration is carried out by comparing the time elapsed between the appearances of the two chemical equivalence points with calibration curves prepared with standard solutions, said time having a well defined functional relationship with the concentration of the sample.

  • Patent number: 4066406
Abstract: A process and apparatus for the voltammetric measurement of sample component quantities is disclosed wherein the material to be measured is passed by means of a pulsating-delivery pump into a voltammetric cell by way of a flow cross-section restriction to the surface of the voltammetric electrode, the liquid jet being adjusted such that its cross-section in the vicinity of the measuring surface of the electrode should be greater than the measurement area of the latter. The obtained voltammetric current curve is integrated at the area below the peak value, the current changing as a function of the pulsating flow rate of the liquid and the material quantities are determined from the integrated data. Preferably, the integration is effected from a value corresponding to 50% of the maximum flow rate to the maximum value and from there again down to 50% of the flow rate maximum.
