    Abstract: The online detection system for type identification and activity measurement of radiations in gas or liquid is provided, which can capture the light signals generated by different radiations in real time, and convert the light signals into the electrical signals, so as to realize the online type identification and activity measurement of the radiations based on the waveforms and the time information of the electrical signals of the different radiations. In addition, based on the characteristic that different radiations have different penetration capabilities, the inner-outer two-layer activity measurement structure is designed, which can discriminate the radiations with the same waveform. Therefore, the present disclosure simplifies the radiation activity measurement process, thereby greatly improving the efficiency of the radiation activity measurement.

    Abstract: An efficient synthesis and assembly method for the large fragment DNA based on the programmable nuclease Argonaute, specifically includes: constructing and treating antibiotic resistance gene reconstructed vectors with linearization, dividing a target DNA into multiple small DNA fragments and then synthesizing the small DNA fragments, followed by loading the synthesized small DNA fragments to the antibiotic resistance gene reconstructed vectors; and the SLIC and resistance gene reconstruction are used to achieve assembly of the target DNA. The method combines the SLIC with a resistance gene reconstruction strategy, allowing for the assembly of 5-6 small fragments in a single resistance gene reconstruction, which is more efficient and time-saving. Moreover, the number of the resistance gene reconstructions can be flexibly chosen according to the length of the DNA fragments. Mutations are not introduced caused by PCR, and the reconstructed large fragment do not need a second sequencing, saving time and costs.

The system generates outlier scores representing the probability that a clinician's prescribing behavior deviates from norms. These scores can be used to evaluate prescription events, generating event scores that indicate the likelihood of illicit activity. The system employs a graph database to model relationships between clinicians, pharmacies, medications, and prescription events. Upon determining that an outlier or event score meets an alert threshold, the system can deny access to electronic prescription applications or alert pharmacies to potential fraud. Reinforcement learning is used to improve model accuracy, minimizing false positives while identifying complex fraud patterns.

    Abstract: The invention discloses Pfu DNA polymerase mutants with reverse transcriptase activity and their applications. The Pfu DNA polymerase mutants include K467R/F588L/W769R, R382H/R385H/V390I, I38L/R97M, E665K/E735K, K118I/N713V, with K118I/N713V showing the strongest reverse transcription activity. These Pfu DNA polymerase mutants still retain the polymerase activity of wild-type enzymes, thus providing higher assurance compared to traditional reverse transcriptase. Meanwhile, these Pfu DNA polymerase mutants exhibit high heat tolerance. Therefore, these Pfu DNA polymerase mutants can efficiently and high fidelity generate cDNA using RNA as a substrate, and further amplify the cDNA under conventional PCR reaction conditions without the need for additional reverse transcriptase, achieving a “one-step” RT-PCR process.
