  • Publication number: 20120156779
Abstract: The present invention relates to a method for cell expansion. In the method, preferably a cell culture product is used, such as a microcarrier, or other adherent cell culture surface, comprising degradable polysaccharide, preferably starch, modified with small molecular weight cell-binding ligands. This allows recovery (detachment) of adhered cells to be aided by degradation of the culture surface with enzymatic agents, such as amylase. The method for cell expansion comprises the following steps: a) adding cells, culture medium and cell culture surface comprising a degradable polysaccharide with guanidine group containing ligands to a bioreactor; b) expanding said cells by adherent cell culture; and c) aiding the detachment of said cells by exposing them to a polysaccharidase to degrade the culturing surface.

  • Publication number: 20110124106
Abstract: A separation device for separating a wanted end product from a liquid sample comprises a container (2) having a first end (5) and a second end (7), the first end having a central orifice (6), a plunger (3) slideably disposed in the container (2) to define a variable liquid receiving chamber between the plunger (3) and the orifice (6), and a permeable partition member (9) mounted to the plunger (3) in a spaced relationship thereto to define a compartment (12) between the partition member (9) and the plunger (3) for receiving liquid density gradient medium (13), wherein liquid may be drawn into the container (2) and expelled therefrom, respectively, through the orifice (6) by movement of the plunger (3) relative to the container (2). A method for separating a wanted end product from a liquid sample is also disclosed.

  • Publication number: 20100120596
Abstract: A separation device adapted for separation of a wanted end product from a sample by centrifugation. According to the invention the device comprises: a first, a second and a third compartment; a first fluid connection between the first and second compartments, arranged to be opened and closed by first closing means; a second fluid connection between the second and the third compartments, arranged to be opened and closed by second closing means. The separation device is adapted to be subjected to centrifugation after having been filled with a first density gradient medium in the first compartment, sample in the third compartment and a second density gradient medium or sample in the second compartment, whereby after centrifugation the wanted end product will settle in the second compartment and the first fluid connection and the second fluid connection will be closed by the first closing means and the second closing means respectively.
