    Abstract: The present application belongs to the field of genetic engineering and medical diagnosis, and particularly relates to serum miRNA markers for diagnosing liver cancer and predicting liver cancer metastasis and a detection kit thereof. The present application discloses the research and development and use of a kit containing three blood miRNAs markers related to human liver cancer. The specific markers are serum-derived miR-122-5p, miR-144-3p and miR-451a. According to the present application, the differential miRNAs are used as the core components of the detection kit, and are combined with corresponding internal references to form the detection kit; and through practical detection application in clinical samples, the reaction conditions and reagent ratios of the kit are optimized, the detection parameters are adjusted, and the standardized process of clinical detection and analysis is preliminarily established, and the kit is used for early diagnosis and metastasis risk assessment of liver cancer.

    Abstract: The present application belongs to the field of biotechnology, and particularly relates to preparation and use of soluble Tim-3 recombinant protein and a mutant protein thereof. The soluble Tim-3 recombinant protein is used to prepare a drug with the function of regulating monocytes or a drug with the function of enhancing tumor immune response, and the amino acid sequence of the soluble Tim-3 recombinant protein is as shown in SEQ ID NO: 1. In addition, the present application further provides the mutant proteins of the sTim-3 recombinant protein and preparation and use of the mutant protein. The mutant recombinant protein is obtained by screening through a directed evolutionary technology, with the function of regulating monocytes or the function of enhancing tumor immune response, and combines with a host codon optimization method to improve the expression efficiency.
