  • Publication number: 20230005568
Abstract: A method to correct amplification bias in amplicon sequencing is disclosed. Amplification efficiency is not constant among different loci in a sample, nor for the same locus in different samples. Differences in 3?-end stability, primer Tm, amplicon length, amplicon GC content, and GC content of amplicon flanking regions all may contribute to amplification bias. Such bias interferes with accurate calculation of copy number for a genomic region of interest and hinders the application of amplicon sequencing for detection of minor copy number variation. The methods of the invention allow correction of amplification bias and enable detection of minor copy number variation using amplicon sequence data.

  • Patent number: 9777431
Abstract: An excess heat recovery apparatus and process for high temperature chlorine dioxide bleaching of pulp is provided. The pulp of the high temperature chlorine dioxide bleaching stage enters a tube-side of a chlorine dioxide preheater through a pipeline. The low-temperature chlorine dioxide in the storage tank enters a shell-side pipeline of the chlorine dioxide preheater. 0.5 mol/L of a stabilizer may be added during preheating to prevent ClO2 from decomposing during the heating process. Preheated chlorine dioxide is then moved into a pulp mixer and the pH is adjusted to 3.2-3.8. The mixed pulp is then moved into a high temperature chlorine dioxide bleaching tower for bleaching. The cooling pulp, now out of the preheater, is washed in an alkaline extraction stage. The waste water from the washing flows directly into an effluent treatment system and is recycled after treatment.

  • Publication number: 20150203810
Abstract: Materials and methods for processing a cell population, such as enriching for a cell of interest from a cell population, are disclosed. The method may include: obtaining a cell population dispersed in a cytocompatible matrix; applying one or more labels to the cell population to distinguish a cell of interest from one or more cells of the cell population; identifying a portion of the cytocompatible matrix containing the cell of interest; and optionally isolating the portion of the cytocompatible matrix containing the cell of interest. The methods may, for example, advantageously provide reduced cell damage or loss relative to other procedures. Also disclosed are devices and compositions that may be useful for performing the methods.

  • Publication number: 20150024476
Abstract: Apparatus and Methods are provided for a microfabricated fluorescence activated cell sorter based on a switch for rapid, active control of cell routing through a microfluidic channel network. This sorter enables low-stress, highly efficient sorting of populations of small numbers of cells (i.e., 1000-100,000 cells). The invention includes packaging of the microfluidic channel network in a self-contained plastic cartridge that enables microfluidic channel network to macro-scale instrument interconnect, in a sterile, disposable format. Optical and/or fluidic switching forces are used alone or in combination to effect switching.

  • Publication number: 20120077191
Abstract: Described herein are compositions and methods useful for the detection of nucleic acid variations. Ligation within a probe or between probes is used to distinguish between probes perfectly complementary to a target and those containing a mismatch. Nucleotide fill-in/extension steps are optionally applied according to the type of assay performed. A circularization and relinearization step can be applied to create a template for further amplification and detection. In certain aspects, portions of a target sequence or its complement are not amplified.
