Abstract: Provided in the present invention is a method for constructing a PacBio sequencing library, comprising the following steps: (1) obtaining a target double-stranded DNA; (2) adding a thermostable RNA ligase to respectively connect two ends of the double-stranded DNA to form a closed loop to obtain a dumbbell-shaped DNA library; (3) purifying the dumbbell-shaped DNA library; and (4) binding with sequencing primers and adding a DNA polymerase to obtain a PacBio sequencing library.

Abstract: A method for quickly extracting long-fragment genomic DNAs by a single reaction tube, comprising the following steps: a) adding a lysis solution and protease K to a sample, to lyse cells and release the genomic DNAs; b) adding a precipitant to the reaction system of step a) to obtain a precipitate of the genomic DNAs; c) washing the obtained precipitate of the genomic DNAs using a washing solution; and d) dissolving the genomic DNAs using a dissolution solution. Also provided is a kit suitable for the method above.
