    Abstract: The present invention describes small molecules that have the activity of directly enhancing insulin sensitivity through epigenetic regulation. These small molecules, therefore, provide a new path for the treatment of type 2 diabetes and insulin resistance in type 1 diabetes or prediabetes and also can be used to treat obesity and chronic liver disease. Methods and compositions including these small molecules or for their administration are described. The methods and compositions can include additional anti-diabetic agents.

    Abstract: The invention provides modulators of the nociceptin receptor (NOP), including both agonists and antagonists. A compound of the invention can be a selective modulator of NOP with respect to the ?- and ?-opioid receptors (MOP and KOP), thus providing a therapeutic method for the treatment of conditions wherein selective NOP modulation is medically indicated and MOP or KOP modulation may be less desirable. A compound of the invention can be a NOP full agonist, partial agonist, inverse agonist, positive or negative allosteric modulator, or a functionally biased agonist.
