Abstract: The present invention relates to a method capable of making one cluster by connecting information of strands generated during a PCR process and tracking the generation order of the generated strands. More specifically, the present invention uses a UID-containing primer so as to enable all parent strands and daughter strands to share one UID, and uses the shared UID so as to connect two strands (parent strand and daughter strand) and furthermore extend to and connect a granddaughter strand, thereby enabling connection to all progeny strands derived from a first copied strand. Accordingly, the present invention is capable of not only making one network (cluster), but also identifying the generation order of strands generated during an amplification process, constructing lineage of amplification, and observing error patterns.

Abstract: Disclosed is a method of preparing an oligonucleotide pool having various combinations of base sequences using one oligonucleotide and next-generation sequencing (NGS). The oligonucleotide pool prepared according to the present invention does not require the use of conventional microarray chips which are expensive and time consuming, and improves efficiency in terms of cost and time by reducing the number of synthesis steps performed to obtain various combinations of oligonucleotides. In addition, according to the method of the present invention, the steps of preparing an oligonucleotide pool can be simplified, the number of combinations of oligonucleotides that can be prepared and utilized can be exponentially increased. Therefore, according to the method of the present invention, applications of genetic materials can be broadly extended.

Abstract: A method for assembling multiple target loci to a single assembled nucleic acid sequence involves assembling the multiple target loci to a single assembled nucleic acid sequence through two polymerase chain reactions (PCRs). A pair of primers for a primary amplification include a target-specific sequence and a 5?-flanking assembly spacer sequence. A primary amplified product amplified by the pair of primers for a primary amplification is assembled to a single shortened nucleic acid sequence in a convenient and easy manner through a set of primers for a secondary amplification, thus enabling the simultaneous detection of multiple target loci. Accordingly, the method and a kit of the present invention may simultaneously detect and analyze multiple variabilities in DNA sequences of a sample, thus remarkably reducing sequencing costs for detecting variabilities, and providing a critical approach and means for achieving the concept of customized medicine.
