  • Patent number: 5587290
Abstract: The invention provides methods and compositions relating to stress tolerant yeast; in particular, yeast mutants deficient in the expression of functional ATH1 gene product (Ath1p). Such yeast have enhanced tolerance to dehydration and freezing, are able to grow to a higher cell density over a range of fermentable carbon source concentrations, are able to produce and/or tolerate higher levels of ethanol, and trehalose. Nucleic acids comprising ATH1 gene sequences are used in hybridization probes and PCR primers, in expression vectors, etc. The invention provides methods for producing a yeast mutant with improved survival ability under stress conditions which involve identifying mutations disrupting ATH1 expression using Ath1-specific reagents or ATH1 hybridization probes or primers.

  • Patent number: 5559010
Abstract: The invention provides methods and compositions for expressing a recombinant gene in eukaryotic cells, especially fungi, preferably yeast. The invention provides transcriptional regulating elements having a novel nucleotide sequence which are capable of trancriptionally regulating the expression of a cis joined gene, typically in response to the availability of certain nutrients to the host cell. Preferred regulatory elements are responsive to nutrient depletion, particulary glucose, ethanol, phosphate or a nitrogen source. Nucleic acid constructs comprising such regulatory elements operably linked to recombinant genes, cells comprising such regulatory elements, and methods of producing recombinant protein in such cells are also provided. The invention discloses regulatory elements which are induced through the ras gene product. Accordingly, the disclosed expression systems also provide a convenient marker for ras gene function.

  • Patent number: 4750747
Abstract: A process of making a seal ring uses an elastic ring having a sealing lip projecting laterally or inwardly about one, innermost edge. The elastic ring is turned inside out so that the one, sealing-lip edge is then outermost before or during fixedly sealing the sealing lip to a supporting ring, the sealing lip having a thin thickness profile effective in this for upsetting the sealing lip. Turning the lipped, elastic ring inside out also crimps a sealing surface of the lipped, elastic ring, which ring bends to engage a shaft axially with the sealing surface, such that lubricant pockets opening from the sealing surface only to the other, free edge of the lipped, elastic ring are uniformly distributed therein circumferentially about the sealing surface. The baffle or side and bottom surface portions of each lubricant pocket merge evenly with each other to form a single ground surface.

  • Patent number: 4595206
Abstract: A seal for a shaft relatively rotatable in an opening in a structure has concentric inner and outer rings. The outer ring is fixed across the opening and has an inwardly-open groove with axial boundary surfaces which converge toward each other outwardly from the axis of the rings. The inner ring is elastic and has two flanges flanking a midsection which receives the shaft for relative rotation therewith. The flanges slidingly bear elastically on the boundary surfaces to produce the seal. The midsection of the inner ring has a larger outside diameter than the inside diameter of the outer ring to prevent extruding the inner ring from the outer ring under rotation or a pressure differential across the seal.

  • Patent number: 4591168
Abstract: A sealing ring for sealing about a relatively-rotatable member is an annular, disk-like ring having a root surface about one edge which is adjacent a flarable, sealing-surface zone for slidingly engaging the member. Torsion ribs project from the root surface. The inside surfaces of the torsion ribs are regular prolongations of the sealing surface and the distance therefrom to opposite, outside surfaces diminishes, preferably continuously to a knife-like edge where the inside and outside surfaces meet. The result peels liquid to be sealed in from the relatively-rotating member so that it does not destructively deform the torsion ribs even at high speeds of relative rotation.

  • Patent number: 4522410
Abstract: An axle or shaft seal has inner and outer sealing rings for respective fixed connection to the axle or shaft and a structure thereabout with respect to which the axle or shaft is relatively rotatable axially. Each of the inner and outer rings has a pair of members associated therewith, at least one sealingly, and extending generally toward the other ring. The members associated with one ring are relatively movable and axially between the members associated with the other ring against which they are urged sealingly by a resilient element therebetween.

  • Patent number: 4522328
Abstract: A shaft seal has an annular housing with a first and a second flange and of a hollow, frustrated cone seal ring, the apex of which is anchored liquid-tight for rotation with axial displaceably but on the shaft. The large end of the conical sealing ring forms a sealing lip which bears elastically on the inner face of the second flange which is directed toward the medium to be sealed, having essentially the shape of the frustum of a hollow. The first flange forms an annular support surface (2) which bears on the seal ring (2) between the apex shaft gripping end and the sealing lip so that the shape of the seal ring is elastically deformed.

  • Patent number: 4474379
Abstract: A radial shaft seal ring has a lip ring for sealing to a shaft for rotation therewith. A U-shaped recess extends around the lip ring. An extension portion of a reinforcing ring which delimits a space on one side thereof to be sealed contacts the U-shaped recess without pressure or connection for low wear relative rotation therein. Preferably, the shoulder of the lip ring defining the side of the U-shaped recess on the sealed-space side of the reinforcing ring has a greater outside diameter than that on the other side to centrifugally aid the seal. More preferably, the latter is done by angling the U-shaped channel to the sealed side to also accommodate axial vibration of the shaft and lip ring.

  • Patent number: 4427205
Abstract: A radial shaft sealing ring is disclosed. The sealing ring comprises a flat circular sealing element fabricated from a polymer material. The outer circumferential region of sealing element is mounted in a holding ring which surrounds the shaft. The sealing element is curved from the outer circumferential region to the inner circumference in the direction toward the sealed off medium with the sealing element being expanded away from the shaft in the region adjacent the inner circumference. The sealing element contacts the shaft intermediate the outer circumferential region and the expanded inner circumference region thereby providing a circular sealing contact area. The sealing element curvature provides an increasing inside diameter for the sealing element along the axial distance of the shaft from the circular sealing contact area to the expanded inner circumference. The inner circumferential area may be provided with swirl fins.
