  • Publication number: 20170313740
Abstract: The present invention provides methods of preparing a peptide having an N-terminal histidine, and compositions comprising a plurality of peptides prepared by the methods. The methods disclosed herein reduce racemization of the N-terminal histidine in the peptides during the synthesis process, thereby improving the yield and purity of the peptide compositions. Exemplary peptides that can be manufactured with the methods include Exenatide, Lixisenatide, and Liraglutide.

  • Patent number: 8658774
Abstract: Meditope variants and methods for their use are provided herein. A meditope variant as described herein comprises a peptide having a sequence CQFDLSTRRLKC (SEQ ID NO:1) or CQYNLSSRALKC (SEQ ID NO:2) that has one or more modifications at of least one amino acid residue of the sequence. Multivalent meditope variant tethering entities are also provided. Such entities may include two or more meditopes coupled via a long linker, multivalent scaffold, biotin-streptavidin, or IgG Fc domain. Further, methods of treating, imaging or diagnosing a disease or condition are provided. Such methods may include administering a therapeutically effective amount of a pharmaceutical composition to a subject, the pharmaceutical compound comprising an antibody-meditope complex; a multivalent tethering agent in combination with a monoclonal antibody or functional fragment thereof; or a combination thereof.

  • Patent number: 6320025
Abstract: Reaction vessels suitable for solid phase polymer synthesis generally comprise: a housing defining a chamber for containing a resin, said housing having an operable inlet port and an operable outlet port in fluid communication with said chamber; and an elongate resin filter having an interior bore, and adapted to enable filtered fluid flow from an exterior inlet surface thereof into the filter interior bore, said filter being disposed in said chamber and having an outlet connection fluidly coupling said filter interior bore to the housing outlet port such that fluid flowing through the housing inlet port into the chamber passes through the filter inlet surface, into the filter interior bore and out the housing outlet port.

  • Patent number: 5453487
Abstract: The invention provides methods and apparatuses for solid-phase peptide synthesis which employ solvent/reagent delivery and measuring systems open to atmospheric pressure coupled to pressurized transfer and reaction systems. The delivery and measuring systems consists of multiple solvent vessels connected to a metering vessel through individual, uninterrupted solvent lines. The metering vessel is connected to a transfer vessel by a valve through which solvent is gravity fed to the transfer vessel. The solvent is now in a closed, single pathway system and can be pressurized from the transfer vessel to a reaction vessel. This closed system allows the complete inversion of the reaction vessel; with the advantage of guaranteeing complete resin to solvent contact without any possibility of loss of resin product.

All of the steps are accomplished by the opening and closing of individual valves that are controlled through a personal computer and system software that allow the user to do these steps individually or in any combination and length.
