    Abstract: A nucleic acid amplification method is disclosed. The method employs a Bw adapter primer having a stem-loop structure to synthesize a complementary strand of a target region, followed by a further synthesis of a complementary strand employing an Fw adapter nucleotide that has a stem-loop structure and an extension-inhibiting modification at the 3?-end. The method makes it possible to simply and specifically form a dumbbell structure in which stem-loop structures are added to both ends of the complementary strand of the target region.
