  • Publication number: 20240392280
Abstract: Disclosed are products and methods for monitoring Klotho protein levels and for stabilizing Klotho protein in a mammalian blood sample, especially at room temperature or without freezing, for a period of time. Methods of detecting and quantifying Klotho protein levels, particularly endogenous and/or exogenous soluble alpha Klotho protein levels, methods of diagnosing Klotho protein deficiency, and methods of increasing Klotho protein levels or production, particularly endogenous and/or exogenous soluble alpha Klotho protein level(s), expression, or production, in a mammalian subject, and products useful in performing the same, including diagnostic kits and compositions for treating Klotho protein deficiency, are disclosed. Compositions are configured or formulated to augment natural soluble alpha Klotho protein production, attenuate Klotho protein damage or degradation, and/or supplement Klotho protein levels with exogenous, recombinant protein.

  • Patent number: 11932676
Abstract: Recombinant Klotho proteins, nucleic acids encoding the same, cell lines and cultures expressing the same, and method of manufacturing and administering the same are disclosed. Proteins have at least 80% amino acid sequence identity to a portion of human alpha Klotho and preferably a solubility or half-life-extending feature such as glycosylation and/or fusion protein tag. Treatment protocols include determining Klotho levels in a subject, calculating a dosage of the protein sufficient to raise the Klotho level in the subject to a predetermined level, administering the dosage to the subject, determining a rate of protein decline in the subject following administration of the protein, calculating a time and/or amount of a subsequent dosage of the protein, and/or administering the subsequent dosage to the subject. Proteins and related treatments for addressing aging-associated and other conditions are disclosed.

  • Patent number: 8005301
Abstract: A method of difference sensing. A first step involves producing a reference image using temporal averaging and spatial averaging. A second step involves producing a test image. A third step involves computationally comparing the reference image with the test image to arrive at a resulting difference image. The temporal averaging and spatial averaging effectively isolates in the difference image coherent changes imbeded in a complex and rapidly changing environment from transient changes inherent to the complex and rapidly changing environment.
