    Abstract: A method of amplifying nucleic acid includes providing a microfluidic device having a space therein. The space is filled with a gel medium. Reagents for carrying out a PCR reaction are supported within the matrix of the gel medium within the space. A nucleic acid containing sample is brought into contact with the gel medium. The sample having whole cells or cell lysate without any prior separation of the nucleic acid from other components of the cells. PCR amplification of nucleic acid from the sample is performed in the space using the PCR reagents.

    Abstract: There is provided a method of inhibiting gene expression by locating a 5?-5 donor splicing site sequence in the 3? UTR of a gene or within coding sequence containing the stop codon of a gene. Stability of homologous mRNA in trans is also adversely affected leading to a reduction in expression. A polynucleotide containing a 5?-donor splicing site sequence in either coding sequence containing the stop codon or the 3? UTR is also provided and in one embodiment is in the form of a vector. The 5?-donor splicing site sequence can be present in multiple copies, for example as a tandem repeat. In one embodiment the 5-donor splicing site sequence has the sequence 5?-MAGGTRAGTA-3? where M is A or C and R is A or G.

    Abstract: Method and therapeutic system in the form of a bandage that administer scopolamine base transdermally in an initial pulse of 10 to 200 .mu.g/cm.sup.2 of skin that quickly brings the concentration of scopolamine in the plasma to a level at which emesis and nausea are inhibited without intolerable side effects, followed by a substantially constant dosage in the range of 0.3 to 15 .mu.g/hr that holds said level. The bandage is a four-layer laminate of, from the top: a protective backing; a gelled, mineral oil-polyisobutene-scopolamine reservoir lamina that is the source of the constant dosage; a microporous membrane that controls the constant dosage rate; and a gelled, mineral oil-polyisobutene-scopolamine adhesive layer that is the source of the pulse dose and the means by which the bandage is attached to the skin.
