  • Patent number: 9605256
Abstract: Procedure for the specific isolation of total DNA content of bacterial germs of different samples, in the course of which the cells are lysated, the DNA content of the lysate is bound selectively, it is washed and then the desalinated linear polymer nucleic acid is eluted from the binding surface in an aqueous solution. Before cell lysis the nonviable bacterial cells are separated from the viable cells on the basis of their different cell surface physical-chemical characteristics, the viable cells of the sample are kept and then lysated using a mechanical and/or enzymatic, favorably lysozyme enzymatic method. After this exclusively double-stranded DNA deriving from the lysate of viable cells is bound on a —SiO2—TiO2— matrix containing chemically activated —OH and dodecylamine groups, and after washing it, the desalinated linear polymer nucleic acid is eluted in an aqueous solution.

  • Publication number: 20130324436
Abstract: Disclosed are procedures and kits for nucleic acid-based molecular diagnostic determination of bacterial germ counts during which procedure evolutionarily conserved genes and genes coding for characteristic pathogenicity markers, favourably microbial enzyme, toxin, special resistance, are detected using real-time PCR amplification method with the application of fluorescent hydrolysis probes. The multiplication of nucleotide chains takes place with oligonucleotides annealing to the structural gene 5? end region and to the adjacent upstream regulatory promoter-operator region so that the presence of the structural gene is shown along with the adjacent upstream regulatory promoter-operator sequences; the functional nature of the structural gene is simultaneously checked. The result is measured with a genome unit equivalent DNA amount calibrated to the germ number of sample units equivalent to standard procedures.

  • Patent number: 5744629
Abstract: The invention relates to improved manufacture of methylene bis-(dibutyl-dithiocarbamate) with ASTM coulour less than 2, by reacting aqueous solutions of dibutyl amine and sodium hydroxide with carbon disulfide, treating the obtained reaction product with methylene dichloride, followed by vacuum stripping, phase separation and filtration of the precipitated sodium chloride. According to the invention, during reaction with carbon disulfide the temperature of the mixture is kept at or below 15.degree. C. and the vacuum stripping is performed in two steps, first between 45.degree. C. and 100.degree. C. and at a pressure of 5-50 kPa, then phase separation is carried out at a temperature between 35.degree. C. and 50.degree. C., followed by stripping the separated organic phase in vacuum also at a temperature between 45.degree. C. and 100.degree. C. and at a pressure of 5-50 kPa.

  • Patent number: 3979259
Abstract: The invention relates to a process for the production of a fermentation broth with increased vitamin B.sub.12 content by a fermentation process carried out with a methane-producing mixed bacterium population under anaerobic septic conditions in the presence of known nutrient components and precursors. According to the invention, methanol, preferably in an amount of 0.5 v/v %, is added for some days, preferably for 3 days to the fermentation broth containing vitamin B.sub.12 used as the starting substance. Thereafter a minor part, preferably 10% of the fermentation broth, is removed and an equal volume of a nutrient broth containing the usual components in tenfold concentrations is added periodically in every 5th to 12th day, then the removal of fermentation broth is interrupted for 0 to 2 days, preferably for 1 day, and only 0.4 to 1.
