Publication number: 20250073209Abstract: RhoA inhibitors that can covalently bind RhoA are described herein. The disclosed RhoA inhibitors show improved inhibitory efficacy (i.e., an IC50 value down to about 400 nM). Further, the disclosed RhoA inhibitors can specifically bind RhoA over other GTPases, such as other Ras GTPases (e.g., K-Ras and N-Ras) and particularly other Rho GTPases (e.g., Cdc42 and Rac1). The combination of high RhoA inhibitory efficacy and high RhoA specificity of the disclosed RhoA inhibitors allow these compounds to show high cytotoxicity and anti-metastatic effects in cancer cells, with negligible toxicity in normal cells, in vitro and in vivo. Pharmaceutical compositions containing the RhoA inhibitors and methods of using the RhoA inhibitors or pharmaceutical composition thereof for treating cancer are also described.Type: ApplicationFiled: August 28, 2024Publication date: March 6, 2025Inventors: Yik Sham Clive Chung, Ying Ki Jason Li, Tin Yan Koo, Hoi Ning Helen Yan

Publication number: 20240296412Abstract: A method includes updating, in a server, a workflow in a contract based on an action by a remote user. The method also includes storing an updated version of the contract in a directory, and providing, to an approver remotely coupled with the server, a link to the updated version. Updating the workflow includes at least one of: selecting a governing law or any other type of field or constraint controlling the updated version of the contract, adding a conditional variable to the contract, the conditional variable configured to acquire a value based on an input from the remote user, or the approver, adding or removing a portion of the contract based on a value of the conditional variable in the contract, and modifying a role for the approver. A system and a non-transitory, computer-readable medium storing instructions to perform the above method are also disclosed.Type: ApplicationFiled: May 10, 2024Publication date: September 5, 2024Inventors: Jason Li, Cai GoGwilt, Mary Zhuang, Angela Kou, Ben Newcomer, Kevin Verdieck

Publication number: 20240265376Abstract: Aspects of the disclosure relate to event processing. A computing platform may prefetch, prior to receipt of a purchase request, a plurality of pay to addresses (associated with an omnibus account, used to route funds from the omnibus account to a merchant account, and not associated with a particular merchant account when prefetched). The computing platform may receive, from a client, a request to make a purchase from a merchant, and may associate the client and the merchant with one of the prefetched pay to addresses/notify the client of the prefetched pay to address. The computing platform may monitor the prefetched pay to address for receipt of a predefined amount of cryptocurrency associated with the purchase. Upon detecting receipt of the predefined amount of cryptocurrency, the computing platform may transfer funds corresponding to the purchase from the omnibus account to an account for the merchant.Type: ApplicationFiled: April 17, 2024Publication date: August 8, 2024Inventors: Dustin Kinsey, Jason Li, Jim Cai

Patent number: 11977334Abstract: A method for determining a wavefront parameter of a patterning process. The method includes obtaining a reference performance (e.g., a contour, EPE, CD) of a reference apparatus (e.g., a scanner), a lens model for a patterning apparatus configured to convert a wavefront parameter of a wavefront to actuator movement, and a lens fingerprint of a tuning apparatus (e.g., a to-be-matched scanner). Further, the method involves determining the wavefront parameter (e.g., a wavefront parameter such as tilt, offset, etc.) based on the lens fingerprint of the tuning apparatus, the lens model, and a cost function, wherein the cost function is a difference between the reference performance and a tuning apparatus performance.Type: GrantFiled: December 28, 2022Date of Patent: May 7, 2024Assignee: ASML NETHERLANDS B.V.Inventors: Duan-Fu Stephen Hsu, Christoph Rene Konrad Cebulla Hennerkes, Rafael C. Patent number: 11880650Abstract: Various aspects of the subject technology relate to systems, methods, and machine-readable media for detecting changes to uploaded documents in a workflow platform. Various aspects may include determining changes between various versions of a media file. Aspects may include detecting what portions of the media file change relative to different instances of the various media file versions. Aspects may also include updating a template element to incorporate the changes to the portions of the media file so that the template element may be used to regenerate a new version of the media file with the incorporated changes being reflected by the template element. Aspects may include determining a new location of text within a portion of the media file that is changed between versions of the media file.Type: GrantFiled: October 19, 2021Date of Patent: January 23, 2024Assignee: Ironclad, Inc.Inventors: Jason Li, Cai Gogwilt

Publication number: 20210393543Abstract: A composition for preventing or treating hypoglycemia in a patient in need thereof has a microgel that includes crosslinked polymers containing glucose-responsive moieties; and blood glucose-raising therapeutic agent loaded on or within the microgel. The microgel further includes any one or a combination of a stabilizing component and a loading-assisting component for the blood glucose- raising therapeutic agent. The polymers containing the glucose-responsive moieties form secondary crosslinks in response to low glucose level, thereby causing shrinking of the microgel and rapid release of the blood glucose-raising therapeutic agent. The composition can be used to prepare a composite microneedle patch for preventing or treating hypoglycemia, where the composition is embedded or integrated within an array of the microneedles of the microneedle patch device.Type: ApplicationFiled: November 8, 2019Publication date: December 23, 2021Inventors: XIAO YU WU, JASON LI, AMIN GHAVAMI NEJAD, BRIAN LU, ADRIA GIACCA
