    Publication date: June 15, 2023
Inventors: Jian-Rong Gao, Caroline Kurtz, Anna Sokolovska, Ning Li
  • Publication number: 20230172997
Abstract: The present disclosure provides recombinant bacteria for production of indole-3-acetic acid (IAA). Pharmaceutical compositions and methods of treating diseases are also included.
    Type: Application
    Filed: May 26, 2021
    Publication date: June 8, 2023
Inventors: Ning Li, Jian-Rong Gao, Christopher George Bergeron
  • Publication number: 20230105474
Abstract: The present disclosure provides recombinant bacterial cells that have been engineered with genetic circuitry which allow the recombinant bacterial cells to sense a patient's internal environment and respond by turning an engineered metabolic pathway on or off. When turned on, the recombinant bacterial cells complete all of the steps in a metabolic pathway to achieve a therapeutic effect in a host subject. These recombinant bacterial cells are designed to drive therapeutic effects throughout the body of a host from a point of origin of the microbiome. Specifically, the present disclosure provides recombinant bacterial cells that comprise a uric acid catabolism enzyme, e.g., a uric acid degrading enzyme, for the treatment of diseases and disorders associated with uric acid, including hyperuricemia and gout, in a subject. The disclosure further provides pharmaceutical compositions and methods of treating disorders associated with uric acid, such as hyperuricemia.
    Type: Application
    Filed: February 25, 2021
    Publication date: April 6, 2023
Inventors: Vincent M. Isabella, Sean Cotton, Jian-Rong Gao, Teodelinda Mirabella, Christopher George Bergeron
  • Publication number: 20220362311
Abstract: The present disclosure provides recombinant bacterial cells that have been engineered with genetic circuitry which allow the recombinant bacterial cells to sense a patients internal environment and respond by turning an engineered metabolic pathway on or off. When turned on, the recombinant bacterial cells complete all of the steps in a metabolic pathway to achieve a therapeutic effect in a host subject. These recombinant bacterial cells are designed to drive therapeutic effects throughout the body of a host from a point of origin of the microbiome. Specifically, the present disclosure provides recombinant bacterial cells comprising a heterologous gene encoding an improved leucine catabolism enzyme with higher activity and/or specificity for leucine over other branched chain amino acids, such as isoleucine or valine.
