    Abstract: Disclosed is an irradiation unit for a flow-cytometry-based analytical instrument and analytical instrument including the same. The irradiation unit includes a light source, a light beam focusing module for focusing the illuminating light beam on an irradiated area, and a light beam shaper disposed between the light source and the light beam focusing module. The light shaper is used for flattening the distribution of light intensity on the irradiated area in a third direction X that is perpendicular to both a first direction in which the light beam spreads and a second direction in which cells to be detected flow. The irradiation unit and the analytical instrument including the same enable the illuminating light beam to form the irradiated area with the uniform distribution of intensity in a predetermined direction to avoid misjudging particles of the same kind entering into different positions of the irradiated area as different, thereby improving accuracy.

    Abstract: An automatic classification method for flow cytometry includes characterizing cells or particles as a vector based on at least two-path optical signals generated when the cells or particles pass through an irradiated area one by one, calculating a distance between the effective cells or particles, in which a shorter distance indicates a higher similarity between the two cells or particles, clustering the cells or particles with high similarity into the same class, continuing to cluster similar cells or particles into the same class until the effective cells or particles are clustered into a number L of classes which should be contained in a sample and is determined based on a measuring principle. This method automatically classifies particles accurately and efficiently.

A sample generation unit includes a gas-liquid transmission controlling module and a flow chamber which are connected to each other, so the sample liquid containing the cells to be detected may flow through the flow chamber while being encircled by the sheath fluid. A signal processing unit receives, converts and processes the scattering light emitted from the flow chamber. The signal processing unit includes at least a photoelectric detection module, wherein the light beam shaping module has at least one aspheric collimating lens and a pair of mutually crossing cylindrical lenses, and the numerical aperture of the aspheric collimating lens is at least 0.3.
