Abstract: Disclosed is a sequencing method, the method comprising providing a solid phase substrate that has a plurality of single-stranded nucleic acids attached to a surface thereof, wherein the 5? ends of the single-stranded nucleic acids are attached to the surface, the single-stranded nucleic acids are polynucleotides that contain an insert-first sequence, the insert is a nucleic acid sequence from a sample to be tested, the first sequence is a preset sequence containing an index-first site, the index is a preset sequence that specifically corresponds to the sample to be tested; providing a first sequencing primer, wherein the sequencing primer can hybridize with the 5? end of the first site; hybridizing the first sequencing primer with the single-stranded nucleic acids and placing same under a condition suitable for polymerized sequencing, so as to determine part of the sequence of the single-stranded nucleic acids by extending the first sequencing primer, so as to obtain a sequencing result.

Abstract: The present application relates to an amino-modified chip, preparation method thereof and use thereof. The amino-modified chip comprises: a substrate and a high polymer, wherein the substrate is modified with epoxy groups; and the high polymer is attached to the substrate via the epoxy groups, at least one structural unit of the high polymer contains an amino, and the amino is a primary amino or a secondary amino.

Abstract: An effective sequencing method, the method comprising: (1) performing first sequencing on a sequencing template on a chip surface, so as to facilitate obtaining first sequencing data by means of first newly generated sequencing strands being formed, the sequencing template being connected onto the chip surface by means of a sequencing adapter; (2) performing first blocking treatment on 3? ends of at least a portion of the first newly generated sequencing strands; and (3) performing second sequencing on the sequencing template, so as to facilitate obtaining second sequencing data by means of second newly generated sequencing strands being formed.
