    Abstract: Disclosed are a plant grain size regulation gene and use thereof, which belong to the technical field of biology. PCFS gene mutants in different crops are obtained by gene editing, and by means of experiments and statistical analysis, it is found that the mutants have a phenotype of enlarged plant grain and part of the mutants have a phenotype of improved stress resistance. The provided gene, mutants, and method for use same are beneficial to increase the crop yield and improve the quality and enhance the resistance of plants to an adverse environment, provide gene resources and technical support for cultivating new plant varieties with large grain weight and high stress resistance, and thereby have important significance and application value for improvement of agronomic traits of crops and breeding of high-yield and high stress-resistant molecules.

    Abstract: Disclosed is a method for increasing expression level of recombinant proteins in Bacillus subtilis through co-expressing Bacillus-derived enhancer factor, belonging to the technical field of enzyme engineering. According to the present disclosure, genes encoding recombinant proteins are respectively co-expressed with Bacillus-derived PonA, PonA truncated forms, OppA or SppA, so as to improve the expression level of the recombinant proteins. The results show that the method provided by the present disclosure can increase the expression level of ultra-high temperature amylase, medium temperature amylase or sucrose isomerase at the shake flask level to 3.96 times, 1.49 times and 2.26 times. The expression level of ultra-high temperature amylase can be increased by 26% in high-density fermentation of a 3L bioreactor. The B. subtilis provides a new developable target as an recombinant protein expression host, and provides technical support for the optimization and modification of the enhancer factors.

The enzyme production of recombinant bacteria can reach 2324 U/mL in a 3.6 L fermenter. The resulting ?-glucosidase TpBgl3A can produce gentioligosaccharides with a high conversion rate using glucose as a substrate at a lower enzyme amount added, which significantly reduces production costs. In the reaction system using glucose and cellobiose as substrates, the conversion rate of gentioligosaccharide reaches 26.2%, which has the potential for industrial production of gentioligosaccharide.
