    Abstract: The present application relates to the technical field of further processing of dairy products, and in particular to a preparation method of milk oligosaccharides, and milk oligosaccharide powder and food prepared thereby. The preparation method comprises the steps of: performing ultrafiltration of whey liquid for at least three times, subjecting the ultrafiltration permeate to nanofiltration concentration for several times, then subjecting the nanofiltration retentate to chromatographic separation and purification, collecting chromatographic collection liquid containing sialyllactose while removing the fraction containing lactose, subjecting the collection to desalination and drying to obtain oligosaccharide powder. The milk oligosaccharides prepared by the present method and the food product containing the same comprise basically bovine milk oligosaccharides, which are light yellow or white in color, light in flavor, uniform in size, and have good thermal stability and solubility.

    Abstract: The present disclosure relates to a method for detecting the contents of whey protein and casein, and/or the ratio thereof in milk powder by liquid chromatography-mass spectrometry. According to the method of the present disclosure, the characteristic peptide segments of whey protein and casein are selected and used for correction, and thus the accuracy and stability of detection of peptide segments by mass spectrometry using external standards are improved; in addition, the method of the present disclosure has the following advantages: easy to pretreat samples, low cost, a wide range of applicability, enable to perform quantitative detection of whey protein at one time, not affected by protein denaturation in the production process, no need to synthesize isotope internal standard, and avoiding influence of ionization efficiency and matrix in mass spectrometry detection by means of their own multiple characteristic peptide segment ratios.

    Abstract: The present invention provides a preparation method for an anti-porcine reproductive and respiratory syndrome cloned pig, comprising: transferring CRISPR/Cas9 targeting vectors and CD163 gene homologous recombination modification vectors into fibroblasts of a pig to obtain positive clone cells, a seventh exon of porcine endogenous CD163 gene being replaced with a eleventh exon of human CD163-L1 gene, so that the positive clone cells are incapable of mediating invasions of PRRSV; taking the positive cell as nuclear transfer donor cells and oocytes as nuclear transfer recipient cells to obtain a cloned embryo by adopting a somatic cell nuclear transfer technology; and impregenating a pig by transferring the cloned embryo into the uterus of the pig, to obtain a cloned pig.
