    Abstract: Provided is a method for trans-differentiating non-neuronal cells of a mammal into neurons. The method comprises: providing a negative regulator capable of reducing the expression of negative regulatory genes, the negative regulatory gene including RCOR1, RCOR2, RCOR3, Sin3a, Sin3b, HDAC1, HDAC2, KDM1A, PHF21A, BAF53a, G9a, USP14, HuR, BrG1, EZH2, CDYL, or HMG20B; and/or providing a positive regulator capable of improving the expression of positive regulatory genes, the positive regulatory genes including DPYSL2, BAF45b, SCF, HuB, HuC, HuD, CYP1B1, or BTRC. An effective amount of the negative regulator or the positive regulator contacting the non-neuronal cells can induce trans-differentiation of the non-neuronal animal cells into neurons.

    Abstract: The transdifferentiation of glial cells into neurons is useful for prevention or treatment of a disease associated with loss of function or death. The present disclosure relates to the field of biomedicines. More specifically, the present disclosure relates to use of a REST inhibitor in the treatment of a disease associated with loss of function or death of neurons. The present disclosure can effectively induce transdifferentiation of astrocytes into dopamine neurons by inhibiting the expression, content or activity of a gene of REST or an RNA thereof or an encoding protein thereof in astrocytes in the brain, and can effectively induce transdifferentiation of Müller glia (MG) into retinal ganglion cells (RGCs) or photoreceptor cells by inhibiting the expression, content or activity of the gene of REST or the RNA thereof or the encoding protein thereof in the retina, thereby preventing and/or treating the disease associated with loss of function or death of neurons.

    Abstract: Provided are an RE1/NRSE element as a target for transdifferentiation of non-neuronal cells into neuronal cells, and the use of an RE1/NRSE blocker in the prevention and/or treatment of diseases associated with loss of function or death of neurons. Also provided is a method for blocking an RE1/NRSE element so as to regulate the expression of a neuron-associated gene in non-neuronal cells. The method comprises reducing binding of REST to an REI/NRSE element or reducing the amount or activity of REST. Also provided is an REI/NRSE element blocker, particularly a binding structural domain of an endogenous RE1/NRSE binding protein and a variant thereof, which can be used for the prevention and/or treatment of diseases associated with loss of function or death of neurons.
