  • Patent number: 12677811
Abstract: A mutated Csf1r gene is disclosed. The mutated Csf1r gene is obtained by changing the 2557th nucleotide of a Csf1r gene from C to A, leading to obtain a mutated protein encoded by the mutated Csf1r gene substitute the 853rd amino acid from proline to threonine. The mutated protein, expression vectors, recombinant viruses, recombinant cells, recombinant bacteria, or recombinant vectors are also disclosed. A method for constructing a murine model with mutations in Csf1r gene. This method includes introducing the targeted vector containing the mutated Csf1r gene into mouse embryonic stem cells, followed by injection into blastocysts to generate F0 generation mice. The F0 generation mice are then bred with mice that specifically express Cre enzyme in tissues, followed by screening. The constructed murine model has significant applications in studying the pathogenic mechanisms of brain diseases caused by microglial cell dysfunction and screening valuable medicine for treating brain diseases.

  • Publication number: 20240336828
Abstract: The present disclosure provides an ultra-high temperature resistant cement slurry system for cementing and the preparation method and use thereof. The cement slurry system comprises cement, an ultra-high temperature strength stabilizer, an ultra-high temperature reinforcing material, a density regulator, an ultra-high temperature suspension stabilizer, a dispersant, a fluid loss additive, a retarder, a defoaming agent and water, wherein the ultra-high temperature suspension stabilizer comprises an ether-based starch, an aluminosilicate and a polyalcohol polymer. The method for preparing the cement slurry system includes dry mixing and wet mixing raw materials homogeneously, respectively, and then homogeneously mixing the dry mix and wet mix to obtain the cement slurry system. The present disclosure further provides use of the cement slurry system for cementing in deep wells and ultra-deep wells at high and ultra-high temperatures.

  • Publication number: 20240058445
Abstract: An inducible chimeric antigen receptor and its application relate to the field of immune therapy. The inducible antigen receptor includes a single-chain variable fragment (scFv) antibody against human CSF1R, a truncated hIgG1 hinge region SH, a T cell co-stimulatory signaling molecule, and a T cell cytoplasmic signaling domain. Transcription of the chimeric antigen receptor is regulated by an inducible expression element tet operator. The induce chimeric antigen receptor molecules can be used to modify T lymphocytes, eliminating the cytokine storm problem of CAR-T cells in the treatment of Alzheimer's disease (AD), and achieving target treatment of AD.
