Procedure for the undifferentiated or myeloid lineage biased expansion of haematopoietic stem cells (HSCs). More specifically, the present invention relates to a procedure of expansion of HSCs from umbilical cord blood, bone marrow or mobilized peripheral blood. Said procedure comprises the steps of expansion culturing of the purified CD34+ cells at constant volume, expansion culturing of said cells at variable volume and the conditioning of the CD34+ cells for transplantation. With this procedure the dose of undifferentiated or myeloid lineage biased HSCs which is necessary for their clinical use is produced reproducibly, robustly and safely.
